摘要
目的 研究内皮细胞白细胞黏附分子 1(ELAM 1)对猪眼小梁细胞形态和肌动蛋白细胞骨架的作用 ,探讨青光眼的发病机制。方法 原代培养的猪眼小梁细胞 ,经白细胞介素 1(IL 1)刺激活化后 ,结合ELAM 1抗体或IgG进一步交联 ,观察细胞形态、细胞间连接的动态改变及肌动蛋白和细胞厚度的改变。结果 未经IL 1作用的猪眼小梁细胞不表达ELAM 1,IL 1作用后猪眼小梁细胞表达ELAM 1。经过IL 1活化的猪眼小梁细胞 ,结合ELAM 1抗体或IgG进一步交联后 ,细胞间隙扩大 ,细胞变圆、变厚 ,细胞立体感增强 ;肌动蛋白变稀疏 ,荧光弥散 ,细胞的刚性降低 ,细胞厚度增加。结论 猪眼小梁细胞能表达ELAM 1,表达的ELAM 1通过与抗体结合或同时通过IgG交联影响肌动蛋白细胞骨架 ,改变细胞形态。
Objective To study the effects of endothelial leukocyte-adhesion molecule-1 (ELAM-1) on the shape and actin cytoskeleton of cultured porcine trabecula r meshwork cells (PTCs). Methods The cultured PTCs were stimulated by interleukin-1(IL-1) and conjugated by ELA M-1 antibody or further cross-linked by IgG. Immunocytochemical procedures were used to study the expression of ELAM-1 and FITC-phalloidin was used to st ain the actin filaments on PTCs; phase contrast microscope was employed to obser ve the dynamic changes of cellular shape and the intercellular conjunction and c onfocal microscope was used to observe the changes of actin and cellular thickne ss.Results IL-1 treatment increased the expression of ELAM-1 in PTCs which resulted in e nlarged intercellular space in PTCs with more rounded and stereoscopic morpholog y after specific antibody conjugation or IgG cross linking. In addition, actin filaments became sparse, cellular rigidity decreased, and the cellular thickness increased after these treatments.Conclusions Our results demonstrated that IL-1 activated PTCs can express ELAM-1 and inte raction of ELAM-1 with specific antibodies and cross linking with IgG, induced a rounding-up of PTCs. This change in cell shape may be due to its action on the actin cytoskeleton.
