摘要
目的 探讨海藻酸作为药物载体在耳科疾病局部治疗中的应用前景。方法 向豚鼠听泡注射藻酸钠与交联剂 ,观察施药后 1~ 2周时中耳黏膜、圆窗膜、咽鼓管及耳蜗组织学改变。结果 未见海藻酸引起鼓室积液。仅见圆窗膜轻度增厚 ,咽鼓管黏膜部分杯状细胞向立方上皮细胞变化 ,但上述部位均未见炎性细胞浸润。中耳黏膜以及耳蜗形态正常。两周内该材料未完全发生降解与排空。结论 藻酸钠凝胶对听泡内组织无致炎性 ,无免疫排斥反应。所见的轻微形态学改变是可逆性的 ,对内耳是安全的。通过对藻酸钠浓度和交联剂的调节 ,可以满足载药后局部应用的需要。
Objective To investigate the prospect of sodium alginate as a drug delivering vehicle in the local application to treat some inner ear diseases.Methods Sodium alginate had been injected into the bullea of guinae pigs together with small amount of Calcium chloride. The morphologic changes in the middle ear mucous membrane, round window membrane(RWM), eustachian tube and cochlear had been observed at the first and second weeks.Results There was slight thickness in the RWM at the 2nd week, and the mucous membrane of eustachian tube changed from goblet cells to cubic cells. However,no inflammatory cells had been found neither in the RMW nor in the eustachian tube, the middle ear mucous membrane and the cochlear appeared normal.Conclusion Sodium alginate causes slight morphologic changes but no inflammation in the bullea. It is relatively safe to the inner ear.
