摘要
采用免疫组织化学S-P法,对成年(2~3月龄)和老年(20~36月龄)Wistar大鼠耳蜗核组织进行ChAT免疫组织化学显色,并利用计算机图像分析系统对免疫反应阳性细胞进行计数。观察老年大鼠耳蜗核组织胆碱乙酰化酶(ChAT)免疫反应的变化,研究其与老年性聋的发病关系。结果发现ChAT免疫反应阳性神经元主要散在分布于背侧核(DCN)的梭型细胞层,老年大鼠耳蜗背侧核ChAT阳性细胞数目少于成年组(P<0.001)。实验结果提示,耳蜗核ChAT阳性细胞数目的减少可能与老年性聋的发病有关。
The purpose of the experiment is to observe the change of choline acetyltransferase (ChAT) immunoreactivity in aged rat cochlear nucleus in order to study its relation with the initiation of presbycusis. The cochlear nucleuses of adult (2~3months) and aged (20~36months) Wistar rats were stained with streptavidin/peroxidase (S P) technique. ChAT immunopositive neurons were counted with a computer aid image analysis system. The result showed that scattered ChAT immunopositive cell bodies were mainly found in the fusiform cell layer of the dorsal cochlear nucleus (DCN). The number of ChAT immunopositive neurons decreased significantly is DCN of aged rat cochlear nucleus ( P <0 001). This indicates that the reduction of ChAT positive neurons in aged rat cochlear nucleus may be related to presbycusis.
