摘要
目的:观察发育和成熟大鼠心室肌细胞中整合素β1表达部位和强度的增龄变化,探讨其生物学意义。方法:选择胎鼠(孕约19d)、新生(9d)、青年(8月)和老年(14月)SD大鼠各7只,用免疫组织化学显色观察心室肌细胞中整合素β1表达,并用计算机图像分析系统定量分析表达强度。结果:各组近心外膜肌细胞中,整合素β1主要表达于胞质内,胎鼠组表达强度与其他组比较具有统计学意义。深层心肌细胞中,胎鼠组主要表达于胞质内,其他组主要表达于胞核和胞膜上,表达强度与青年和老年组相比具有统计学意义。结论:大鼠心肌细胞发育和成熟过程中,近心外膜心室肌细胞整合素β1分布一致,而深层心室肌细胞整合素β1分布由细胞质转变为细胞核和细胞膜。
Objective: To observe the distributive and intensive changes of integrin β1 in ventricular myocytes during developing and mature rat, and to probe the biological significance. Methods: SD rats were selected, and divided into four groups: fetal group (pregnant about 19 d), newborn group (9 d), young group (8 months) and old group (14 months). The expressive distribution and intensity of integrin βl was detected by immunohistochemistry. The expression intensity of integrin β1 was quantitativly analyzed with computer image analysis system. Results: In myocytes neighboring epicardium, the expression of integrin β1 was mainly located int cytoplasm in every group. The expressive intensity in the fetal group was significantly different from that in other groups. In deep myocardium, the expression was mainly located in cytoplasm in the fetal group, and in cellular nucleus and membrane in other groups. The expressive intensity in the fetal group was significantly different from that in the young and old groups. Conclusion: During the development of mouse, integrin β expressed in the ventricular myocytes neighboring the epicardium is unchanged, while the integrin β expressed in the deep ventricular myocytes changes from the cytoplasm to nucleus and membrane.
