摘要
目的:探讨大鼠海马CA3区胆碱能神经元增龄性变化的规律.方法:SD大鼠随机分为1~2月龄、4~5月龄,11~12月龄、≥24月龄4组,常规石蜡包埋海马连续冠状切片,尼氏染色、乙酰胆碱转移酶(ChAT)免疫组织化学显色,图像分析仪测量ChAT免疫反应阳性产物的光密度及其阳性神经元的多种形态学参数.结果:随着年龄的增长,大鼠海马CA3区ChAT免疫反应阳性产物的光密度及其阳性神经元多种形态学参数随增龄而减小.结论:海马CA3区胆碱能神经元随年龄增长而逐渐出现衰老性变化.
Objective: To investigate the changs of ChAT-neurons in the CA3 area of rat's hippocampus with aging. Methods: Sprague-Dawley rats were randomly divided into 4 groups: 1-2 months group, 4-5 months group, 11-12 months group and 24 months group. Routine paraffin was made for Nissl staining and choline acetyltransferase (CHAT) immunohistochmical staining. Then immtmoreactivity were measured with color medical image analysis system Results: The average opitical densities of immunoreactive products in CA3 area of the hippocampus were significantly lower. The important morphological parameters such as the volume density, and the numerieal density of the ChAT-positive neurous decreased with increasing of age. Conclusion: The expression of the ChAT-positive neurons in CA3 area of rat's hippocampus may be gradually decreasing with aging.
