摘要
目的探讨乳移平对乳腺癌细胞迁移能力及mTOR信号通路的影响。方法对我院95例乳腺癌患者,取术后癌变组织,分离乳腺癌细胞,RT-PCR、Western blot法测mTOR水平及蛋白状况,流式细胞术、MTT法检测细胞的凋亡、增殖状况,Transwell检测乳腺癌细胞迁移能力。结果RT-PCR检测mTOR水平结果显示,与Nc组比对,R2组mTOR水平有所降低,R4组mTOR水平较R2组低,R6组mTOR水平最低,mTOR水平随药物浓度增加而降低(均P<0.05)。Western blot法检测mTOR蛋白发现,与Nc组比对,R2组mTOR蛋白有所降低,R4组mTOR蛋白低于R2组,R6组mTOR蛋白最低,mTOR蛋白表达随药物浓度增加而减少(均P<0.05)。流式细胞术检测发现,与Nc组比对,R2组乳腺癌细胞少量凋亡,R4组乳腺癌细胞凋亡数量较R2组多,R6组凋亡细胞最多,乳腺癌细胞凋亡岁药物浓度增加而升高(均P<0.05)。MTT检测结果显示,与Nc组比对,R2组细胞增值能力减弱,R4组增值能力弱于R2组,R6组增值能力最弱,乳腺癌细胞增值人能力随药物浓度增加逐渐下降(均P<0.05)。Transwell迁移实验发现Nc组癌细胞迁移能力强,显微镜下细胞数量多,R2组癌细胞数量较Nc组少,迁移能力较弱,R4组迁移能力弱于R2组,R6组细胞迁移能力最弱(均P<0.05)。结论乳移平对乳腺癌细胞迁移能力有抑制作用,降低mTOR信号通路在乳腺癌细胞中的活性。
ObjectiveTo investigate the effect of breast translation on the migration ability of breast cancer cellsand m TOR signaling pathway.MethodsNinety-five patients with breast cancer were enrolled in this study.Breast cancercells were isolated after operation.RT-PCR and Western blot were used to detect m TOR level and protein.Flow cytometryand MTT were used to detect cell apoptosis and proliferation.Transwell method can determine the invasive ability of breastcancer cells.ResultsThe level of m TOR detected by RT-PCR showed that compared with Nc group,the level of m TOR inR2 group was lower,that in R4 group was lower than that in R2 group,and that in R6 group was the lowest.The level ofm TOR decreased with the increase of drug concentration(all P<0.05).Western blot analysis of m TOR protein showed thatcompared with Nc group,m TOR protein in R2 group decreased,m TOR protein in R4 group was lower than that in R2 group,m TOR protein in R6 group was the lowest,and m TOR protein expression decreased with the increase of drug concentration(all P<0.05).Flow cytometry showed that compared with Nc group,breast cancer cells in R2 group had a small amount ofapoptosis,while breast cancer cells in R4 group had more apoptotic cells than those in R2 group,and apoptotic cells in R6 group had the most apoptotic cells.Drug concentration of apoptotic age of breast cancer cells increased(all P<0.05).MTTresults showed that compared with Nc group,the cell proliferation ability of R2 group was weaker,that of R4 group wasweaker than that of R2 group,that of R6 group was weakest,and that of breast cancer cell proliferation ability graduallydecreased with the increase of drug concentration(all P<0.05).Transwell migration experiment showed that the migrationability of cancer cells in Nc group was strong,the number of cells under microscope was large,the number of cancer cells inR2 group was less than that in Nc group,the migration ability of cancer cells in R4 group was weaker than that in R2 group,and the migration ability of cancer cells in R
