肝癌组织中TGF-β1、β-catenin和EMT相关蛋白表达水平及临床意义 被引量:8

摘要 目的:探讨肝癌组织样本中转化生长因子1(TGF-β1)、β-catenin和上皮间质转化(EMT)相关蛋白表达水平与临床病理学指标的相关性。方法:采用免疫组织化学法检测TGF-β1、β-catenin、E-cadherin及vimentin在62例肝癌患者组织样本中的表达水平,分析TGF-β1与肝癌患者的临床病理学相关性。结果:62例肝癌组织样本中48.39%(30/62)高表达TGF-β1,高表达TGF-β1肝癌组织样本中同时表现出β-catenin高表达、E-cadherin表达降低和Vimentin表达增加。高表达TGF-β1与E-cadherin的表达呈负相关,和β-catenin和Vimentin的表达呈正相关。根据免疫组化评分组织样本分为高表达TGF-β1组(n=30)和低表达TGF-β1组(n=32)。TGF-β1的高表达与肿瘤大小、是否有血管侵犯、AFP水平、肿瘤数目和分化程度具有显著相关性。结论:肝癌组织中高表达TGF-β1伴随着β-catenin表达增加,E-cadherin表达降低,Vimentin表达增加,提示体内随着TGF-β1含量的升高,肝癌细胞可能通过Wnt/β-catenin信号通路诱导肝癌细胞发生EMT样改变,发生侵袭转移的潜能随之增加,TGF-β1可能成为未来临床上进行肝癌干预治疗一个新靶点。 Objective:To detect the expression of transforming growth factor-β1(TGF-β1),β-catenin and Epithelial-mesenchymal transition(EMT)-related proteins in HCC and determine the relationship with their clinicopathological data.Methods:Using Immunohistochemistry test to identify the expression of TGF-β1,β-catenin,E-cadherin and vimentin in the liver cancer tissue and then according to the stainning degree of TGF-β1,we divided 62 patients into high and low expression group and then analyzed their date withβ-catenin and EMT-related protein and clinicopathological data.Results:TGF-β1 expression mainly located in liver cancer cell cytoplasm and high expressed in 48.39%(30/62)HCC samples.We also found that CCL20 expression was accompanied with decreased expression of E-cadherin and increase expression ofβ-catenin and vimentin.62 patients were divided into high TGF-β1 expression group(30/62)and low expression group(32/62)and high TGF-β1 expression was negatively correlated with the expression of E-cadherin(4/30,13.33%,P<0.001)and positively correlated with the expression ofβ-catenin and Vimentin(29/30,96.67%,27/30,90%,P<0.001).Further study with the patient's pathology and clinical data,we inferred that the TGF-β1 expression was significantly correlated with tumor size(P=0.000),vascular invasion(P=0.014),AFP level(P=0.043),tumor number(P=0.008)and differentiation degree(P=0.007).Conclusion:TGF-β1 expressed was associated with increased expression ofβ-catenin and Vimentin,decreased expression of E-cadherin.These results suggest that TGF-β1 might induce liver cancer cells present EMT-like changes by Wnt/β-catenin signaling pathway,TGF-β1 may become an new target of HCC clinical intervention in future.
