摘要
目的研究胃癌组织中长链非编码RNA(lncRNA)心机梗塞相关转录因子(MIAT)的表达水平及临床意义.方法使用qRT-PCR法检测92例胃癌患者的胃癌组织及癌旁正常组织中lncRNA MIAT的表达水平,并分析其与胃癌患者临床病理学指标的相关性;分析胃癌患者的生存情况及影响其预后的独立危险因素.结果lncRNA MIAT在胃癌组织中的表达水平明显高于癌旁正常组织,且Ⅲ~Ⅳ期高于Ⅰ~Ⅱ期(P<0.05).lncRNA MIAT高表达组与lncRNA MIAT低表达组的肿瘤分期、淋巴结转移情况有显著差异(P<0.001).lncRNA MIAT高表达组的术后5年生存率明显低于lncRNA MIAT低表达组(P<0.05).肿瘤分期、淋巴结转移及lncRNA MIAT表达水平为影响胃癌患者生存情况的独立危险因素(P<0.05).结论lncRNA MIAT高表达可提示患者预后不良.lncRNA MIAT可作为胃癌诊断分子标志物与潜在治疗靶点,在胃癌临床预后具有重要的医学意义.
Objective To study the expression level and clinical significance of long non-coding RNA(lncRNA)myocardial infarction-associated transcript(MIAT) in gastric cancer. Methods The expression level of lncRNA MIAT in gastric cancer tissues and paracancerous normal tissues of 92 patients with gastric cancer were measured by qRT-PCR method, and its correlation with clinical pathological indexes of gastric cancer patients were analyzed. The survival condition of gastric cancer patients and the independent risk factors affecting the prognosis were analyzed. Results The expression level of lncRNA MIAT in the gastric cancer tissues was significantly higher than that in the paracancerous normal tissues, and that in the stage Ⅲ-Ⅳ was higher than the stage Ⅰ-Ⅱ(P<0.05). There were significant differences in tumor stage and lymph node metastasis between the lncRNA MIAT high expression group and the lncRNA MIAT low expression group(P<0.001).The five-year survival rate of the lncRNA MIAT high expression group was significantly lower than that of the lncRNA MIAT low expression group(P<0.05). Tumor stage, lymph node metastasis and expression level of lncRNA MIAT were independent risk factors for survival of patients with gastric cancer(P<0.05). Conclusion High expression of lncRNA MIAT can indicate poor prognosis of patients. lncRNA MIAT can be used as a molecular marker and potential therapeutic target in the diagnosis of gastric cancer, and has important medical significances in the clinical prognosis of gastric cancer.
