摘要
目的分析不同年龄健康人群血小板中C反应蛋白(CRP)、肿瘤坏死因子(TNF)α、白细胞介素10(IL-10)的表达水平、变化规律及与血浆同型半胱氨酸(Hcy)的相关性。方法选取192例30(30~39)、40(40~49)、50(50~59)、60(60~69)、70(70~79)岁年龄段的健康体检人群血液标本,提取EDTA-K2抗凝全血中的血小板,采用酶联免疫吸附试验(ELISA)检测血小板中炎症标志物CRP、TNF-α和IL-10的蛋白水平,同时对血浆中Hcy水平进行分析。结果年龄较高组(如60,70岁组)血小板中CRP、TNF-α含量均明显高于年龄较低组(如30,40,50岁组)(P<0.05),而IL-10含量年龄较低组(如30,40岁组)明显高于年龄较高组(如60,70岁组)(P<0.01);血浆Hcy水平随年龄增长而增高(P<0.05)。Pearson相关分析表明,血浆Hcy水平与血小板中CRP、TNF-α含量呈正相关,与IL-10呈负相关(P<0.001)。结论血小板中炎症因子紊乱与衰老密切相关,为探讨血小板在衰老中的作用提供了依据,可作为新型衰老指标。
Objective To analyze the expression levels and patterns of C-reactive protein(CRP),tumor necrosis factor(TNF)-αand interleukin-10(IL-10)in the platelets of healthy people of different ages and their correlation with plasma homocysteine(Hcy)levels.Methods Totally 192 healthy people in their 30(30-39),40(40-49),50(50-59),60(60-69),and 70(70-79)years were studied.Platelets from EDTA-K2 anticoagulated whole blood were extracted and the protein levels of inflammatory markers CRP,TNF-αand IL-10 in platelets were detected by enzyme-linked immunosorbent assay(ELISA).Hcy levels in plasma were also analyzed.Results The platelet CRP and TNF-αlevels in the older groups(such as the 60,and 70-year-old groups)were significantly higher than those in the younger groups(such as the 30,40,and 50-year-old groups)(P<0.01,P<0.05),while the levels of IL-10 in the younger groups(such as the 30,and 40-yearold groups)were significantly higher than those in the older groups(such as the 60,70-year-old groups)(P<0.01).Hcy levels in plasma increased with age(P<0.05).Pearson correlation analysis showed that plasma Hcy levels were positively correlated with platelet CRP and TNF-αlevels,but negatively with IL-10 levels(P<0.001).Conclusion The disorder of inflammatory cytokines in platelets is closely related to aging,which provides a clue to explore the role of platelets in aging and can be used as a new type of aging indicator.
