摘要
目的 应用现代化代谢组学前沿技术探索中药沙棘抗衰老的作用机制,为筛选延缓衰老的药物提供实验依据.方法 将SD大鼠分为4月龄组(n=10)、24月龄组(n=11)和沙棘干预24月龄组(n=12).收集各组大鼠尿液进行1H NMR分析.应用Hotelling's T2和趋势过滤方法分析各组尿液中代谢物,观察中药沙棘对于代谢物的影响.结果 鉴定13个与衰老有关的代谢物,中药沙棘干预组代谢谱发生明显改变.结论 中药沙棘可通过改善部分代谢途径延缓衰老,为抗衰老药物的筛选提供了新的实验方法和理论依据.
Objective To explore the anti-aging mechanism of Hippophae rhamnoides L using metabonomics in order to provide experimental data for drug development on anti-aging. Methods SD rats were divided into three groups: 4 months group, 24 months group, Hippophae rhamn0ides L treated group (24 months). Urine of each rat was collected for metabolite analysis by 1H NMR based on the method from Hotelling' s T2 and method of trend filtering algorithm. Results The data showed that Hippophae rhamnoides L changed the metabolism spectrum of rat by identifying and analyzing 13 metabolite related to aging. Conclusion Hippophae rhamnoides L can delay the process of aging by changing the metabolism, which may provide a new theory and way for drug development against aging.
