摘要
目的探究食管鳞癌及癌旁组织中长链非编码核糖核酸DLX6-AS1(LncRNA DLX6-AS1)的表达水平及其与预后的关系。方法收集佳木斯市中心医院2013年10月-2015年4月期间收治的56例食管鳞癌患者的术后癌组织及癌旁组织。采用实时定量聚合酶链锁反应(qRT-PCR)检测LncRNA DLX6-AS1在不同组织中的相对表达量,术后对患者随访3年,分析其表达情况与患者临床病理特征及预后之间的关系。结果LncRNA DLX6-AS1在癌组织中相对表达量(2.78±0.13)明显高于癌旁组织(1.03±0.08),差异有统计学意义(P<0.05)。LncRNA DLX6-AS1在食管鳞癌癌组织中的表达水平与患者年龄、性别、肿瘤位置均无关(P>0.05),但与患者临床分期、肿瘤分化程度及淋巴结转移情况有关(P<0.05)。Kaplan-Meier分析显示LncRNA DLX6-AS1高表达患者的生存率及平均生存期[21.21%,(23.42±1.66)月]显著低于低表达组[47.82%,(30.13±1.50)月,P<0.05],COX多元回归分析结果显示LncRNA DLX6-AS1、淋巴结转移为影响患者生存率的独立危险因素。结论LncRNA DLX6-AS1在癌组织中相对表达量显著上调,且与临床病理特征及预后密切相关,预测LncRNA DLX6-AS1可能是食管鳞癌新的治疗靶点。
Objective To investigate the expression of long non-coding ribonucleic acid DLX6-AS1(LncRNA DLX6-AS1)in esophageal squamous cell carcinoma and adjacent tissues and its relationship with prognosis.Methods 56 cases of esophageal squamous cell carcinoma(ESCC)were collected from October 2013 to April 2015.Real-time quantitative polymerase chain reaction(qRT-PCR)was used to detect the relative expression of LncRNA DLX6-AS1 in different tissues.The patients were followed up for three years,and the relationships between LncRNA DLX6-AS1 expression with clinicopathological characteristics and prognosis were analyzed.Results The relative expression of LncRNA DLX6-AS1 in cancer tissues was(2.78±0.13),which was significantly higher than(1.03±0.08)in adjacent tissues(P<0.05).The expression level of LncRNA DLX6-AS1 in esophageal squamous cell carcinoma tissues was not related to the age,sex and location of the tumor(P>0.05),but was related to the clinical stage,the degree of differentiation and lymph node metastasis(P<0.05).Kaplan-Meier analysis showed that the survival rate and mean survival time of patients with high expression of LncRNA DLX6-AS1 were 21.21%and(23.42±1.66)months,which were significantly lower than 47.82%and(30.13±1.50)months of patients with low expression of LncRNA DLX6-AS1(P<0.05);COX multiple regression analysis showed that LncRNA DLX6-AS1 and lymph node metastasis were independent risk factors for survival.Conclusion The relative expression of LncRNA DLX6-AS1 in esophageal squamous cell carcinoma was significantly up-regulated,which was closely related to clinicopathological features and prognosis.LncRNA DLX6-AS1 might be a new therapeutic target for esophageal squamous cell carcinoma.
