丹皮酚联合甲氨蝶呤对类风湿关节炎大鼠的改善效果 被引量:1

Improvement effect of paeonol combined with methotrexate in rats with rheumatoid arthritis

摘要 目的探究丹皮酚联合甲氨蝶呤对类风湿关节炎大鼠炎症反应和免疫功能的影响。方法选取健康SPF级雄性SD大鼠50只,按照随机数字表法将大鼠分为五组,每组10只。分组如下:对照组、模型组、丹皮酚组、甲氨蝶呤组、联合组。模型组、丹皮酚组、甲氨蝶呤组、联合组均建立类风湿关节炎大鼠模型。检测大鼠处理21 d后足厚度、足周长、关节炎指数。ELISA检测各组大鼠血清中炎症因子如白介素-1β(IL-1β)、白介素-4(IL-4)、白介素-10(IL-10)、白介素-17A(IL-17A),免疫因子指标如白介素-2(IL-2)、肿瘤坏死因子-α(TNF-α)和肝功能指标天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)含量。结果与对照组大鼠相比,其余各组大鼠足厚度、足周长、关节炎指数均出现不同程度的升高(F=52.820、42.060、314.000),血清中IL-1β、IL-17A、IL-2、TNF-α、AST和ALT含量均显著升高(F=111.300、282.900、72.980、86.200、28.250、82.430),IL-4和IL-10含量均显著降低(F=90.420、94.650),差异均有统计学意义(P均<0.05)。与模型组大鼠相比,丹皮酚组、甲氨蝶呤组和联合组大鼠足厚度、足周长、关节炎指数均显著降低,血清中IL-1β、IL-17A、IL-2和TNF-α含量显著降低,IL-4和IL-10含量显著升高,甲氨蝶呤组和联合组大鼠AST和ALT含量显著升高,差异均有统计学意义(P均<0.05)。与丹皮酚组、甲氨蝶呤组相比,联合组大鼠足厚度、足周长、关节炎指数均显著降低,血清中IL-1β、IL-17A、IL-2和TNF-α含量显著降低,IL-4和IL-10含量显著升高,差异均有统计学意义(P均<0.05)。与甲氨蝶呤组相比,联合组大鼠AST和ALT含量显著降低,差异均有统计学意义(P均<0.05)。结论丹皮酚联合甲氨蝶呤联合用药相比单独用药,对大鼠类风湿关节炎大鼠症状、炎症反应改善更为明显。联合用药能明显改善大鼠免疫性炎症反应,能显著降低甲氨蝶呤单独用药导致的肝损伤� Objective To investigate the effects of paeonol combined with methotrexate on inflammatory response and immune function in rats with rheumatoid arthritis.Methods 50 healthy SPF male SD rats were randomly divided into 5 groups with 10 rats in each group.The groups were as follows:control group,model group,paeonol group,methotrexate group and combined group.A rat model of rheumatoid arthritis was established in the model group,paeonol group,methotrexate group and combined group.The foot thickness,foot circumference and arthritis index were measured after 21 days of treatment.The contents of inflammatory factors such as interleukin-1β(IL-1β),interleukin-4(IL-4),interleukin-10(IL-10),interleukin-17A(IL-17A),immune factor indexes such as interleukin-2(IL-2),tumor necrosis factor-α(TNF-α)and liver function indicators such as aspartate aminotransferase(AST),alanine aminotransferase(ALT)in serum were detected by ELISA.Results Compared with the control group,the foot thickness,foot circumference and arthritis index of other groups increased in varying degrees(F=52.820,42.060,314.000),the contents of IL-1β,IL-17A,IL-2,TNF-α,AST and ALT in serum increased significantly(F=111.300,282.900,72.980,86.200,28.250,82.430),and the contents of IL-4 and IL-10 decreased significantly(F=90.420,94.650),the differences were statistically significant(all P<0.05).Compared with the model group,the foot thickness,foot circumference and arthritis index of the paeonol group,methotrexate group and combined group were significantly reduced,the contents of IL-1β,IL-17A,IL-2 and TNF-α in serum were significantly reduced,the contents of IL-4 and IL-10 were significantly increased,and the contents of AST and ALT in methotrexate group and combined group were significantly increased(all P<0.05).Compared with paeonol group and methotrexate group,the foot thickness,foot circumference and arthritis index of the combined group decreased significantly,the contents of IL-1β,IL-17A,IL-2 and TNF-α in serum decreased significantly,and the contents of
