摘要
目的探讨脂肪与肥胖相关基因(FTO)与原发性膝关节骨关节炎(PKOA)的相关性。方法前瞻性选取2019年1月-2020年1月广东省湛江中心人民医院收治的150例PKOA患者作为病例组,选择同期体检的80例作为对照组,记录两组患者基线资料,血清炎症水平、FTO蛋白浓度以及淋巴细胞中FTO mRNA的表达水平;同时根据WOMAC量表评分将病例组患者分为轻度组(45例)、中度组(55例)、重度组(50例),对比FTO蛋白浓度以及淋巴细胞中FTO mRNA在不同严重程度中的表达水平,采用Pearson相关分析FTO与PKOA间的相关性。结果病例组和对照组BMI、FTO/GAPDH、FTO蛋白、TNF-α、IL-6、IL-1β对比差异具有统计学意义(P<0.05);轻度组、中度组和重度组FTO/GAPDH、FTO蛋白对比差异具有统计学意义(P<0.05),Pearson相关性分析显示:FTO/GAPDH、FTO蛋白水平与PKOA呈显著正相关(r=0.886;r=0.914)(P<0.05)。结论FTO与PKOA显著相关性,可参与PKOA的发生和发展。
Objective To explore the relationship between fat and obesity-related genes(FTO)and primary knee osteoarthritis(PKOA).Methods Prospectively selected 150 PKOA patients admitted to our hospital from January 2019 to January 2020 as the case group,and selected 80 patients undergoing physical examination during the same period as the control group.Record baseline data of the two groups of patients,serum inflammation level,FTO protein concentration,and FTO mRNA expression level in lymphocytes;At the same time,patients in the case group were divided into mild group(45 cases),moderate group(55 cases),and severe group(50 cases)according to the WOMAC scale score.The FTO protein concentration and the FTO mRNA in lymphocytes were compared in different degrees of severity.Expression level,Pearson correlation was used to analyze the correlation between FTO and PKOA.Results There were statistically significant differences in BMI,FTO/GAPDH,FTO protein,TNF-α,IL-6,and IL-1β between the case group and the control group(P<0.05).The difference of FTO/GAPDH and FTO protein in mild group,moderate group and severe group was statistically significant(P<0.05);Pearson correlation analysis showed that FTO/GAPDH and FTO protein levels were significantly positively correlated with PKOA(r=0.886;r=0.914)(P<0.05).Conclusion FTO has a significant correlation with PKOA and can participate in the occurrence and development of PKOA.
