摘要
目的:研究会泽晾晒烟中的化学成分及其细胞毒活性。方法:运用AB-8大孔树脂柱色谱、硅胶柱色谱、MCI柱色谱、Sephadex LH-20凝胶柱色谱及半制备型高效液相色谱等方法和MS、NMR等波谱法以及理化性质对化合物进行分离、纯化和鉴定。采用CCK8法测定化合物对人肝癌SMMC-7721细胞的细胞毒活性。结果:从会泽晾晒烟95%乙醇提取物中分离得到24个化合物,分别鉴定为:可替宁(1)、莨菪亭(2)、吲哚-3-羧酸(3)、(1H-indol-3-yl) oxoacetamide(4)、2′-O-甲基腺苷(5)、降烟碱(6)、尼古丁(7)、3-羟基-5α,6α-环氧-β-紫罗兰酮(8)、山柰酚(9)、金雀异黄酮(10)、烟酰胺(11)、麦斯明(12)、绿原酸(13)、2-甲基喹啉(14)、3,4-二羟基苯甲酸(15)、香草酸(16)、新烟碱(17)、儿茶素(18)、芸香叶苷(19)、凯林(20)、3,4-二羟基肉桂酸(21)、二甲基异咯嗪(22)、4-羟基苯乙醇(23)、咖啡酸苯乙酯(24)。结论:其中,化合物4、5、8、22为首次从该属植物中分离得到,化合物3、4、9~11、18~22表现出一定的细胞毒活性。
Objective: To study the chemical constituents and cytotoxic activity of Nicotiana tabacum in Huize county.Methods: The AB-8 macroporous resin column chromatography, silica gel column chromatography, MCI column chromatography, Sephadex LH-20 gel column chromatography, semi preparative high performance liquid chromatography, MS, NMR spectroscopy and physicochemical properties were used to isolate, purify and identify the compounds.The cytotoxic activity of the compounds against SMMC-7721 cells was determined by CCK8 assay.Results: Twenty-four compounds were isolated from 95%ethanol extract of Nicotiana tabacum in Huize county and identified as cotinine(1),scopoletin(2),indol-3-carboxylic acid(3),(1 H-indol-3-yl) oxoacetamide(4),2′-O-methyladenosine(5),nornicotine(6),nicotine(7),3-hydroxy-5α,6α-epoxy-β-ionone(8),kaempferol(9),genistein(10),nicotinamide(11),myosmine(12),chlorogenic acid(13),2-methylquinoline(14),3,4-dihydroxybenzoic acid(15),vanillic acid(16),anabasine(17),catechin(18),rutin hydrate(19),khellin(20),3,4-dihydroxycinnamic acid(21),lumichrome(22),4-hydroxyphenethyl alcohol(23),caffeic acid phenethyl ester(24),respectively.Conclusion: Among them, compounds 4,5,8 and 22 are isolated from Nicotiana genus for the first time, compounds 3,4,9 to 11,18 to 22 show definite antitumor activities.
