加味当归芍药散对Con A诱导的免疫性肝损伤小鼠保护作用 被引量:7

Protective Effects of Modified Danggui Shaoyao San on Con A-Induced Immunological Liver Injury in Mice

摘要 目的:考察加味当归芍药散对刀豆蛋白A(Con A)诱导的免疫性肝损伤小鼠的保护作用,并探讨其作用机制。方法:将30只BALB/c雄性小鼠随机分为5组:空白对照组、Con A模型对照组、联苯双酯0.2 g/kg组、当归芍药散5 g/kg、15 g/kg组。每组6只。灌胃给予相应药物或生理盐水,连续7日,末次药后1 h,采用Con A制备急性肝损伤模型,造模8 h后取材。采用试剂盒检测肝脏组织匀浆液SOD及血清ALT、AST的活力;ELISA法检测肝脏匀浆液MDA及血清NO、TNF-α、IFN-γ、IL-4及IL-6的含量;HE染色法观察肝脏组织病理学的改变;应用Western Blot法检测肝脏组织中Nrf2、Keap1蛋白的表达情况。结果:与空白对照组比较,Con A模型对照组小鼠肝脏匀浆液中MDA含量升高,而SOD活性降低,血清中ALT、AST、NO、TNF-α、IFN-γ、IL-4及IL-6水平显著升高。加味当归芍药散15 g/kg能显著降低肝脏匀浆液MDA含量及增强SOD活性,加味当归芍药散5、15 g/kg均可抑制血清中ALT、AST、NO、TNF-α、IFN-γ、IL-4及IL-6含量的升高,明显上调Nrf2、Keap1蛋白的表达。结论:加味当归芍药散对Con A诱导急性小鼠肝损伤具有明显的保护作用,其机制可能与上调Nrf2信号通路有关。 Objective:To investigate the protective effects of modified Danggui-Shaoyao-San(mDSS)on immunological liver injury induced by concanavalin A(Con A)and to explore its mechanism.Methods:Thirty male BALB/c mice were randomly divided into five groups,the control group,Con A model group,0.2 g/kg biphenyl diester group,5 g/kg mDSS group and 15 g/kg mDSS group,with each group consisted of six mice.Mice were administered with corresponding drugs or saline water once a day,for constantly 7 days.1 hour after the last administration,Con A was used to establish acute liver injury model.8 hours after modeling,the kits were used to detect the activities of SOD,ALT and AST in liver tissue homogenate and serum,ELISA was used to detect the contents of MDA,NO,TNF-α,IFN-γ,IL-4 and IL-6 in liver homogenate and serum.Pathological changes of liver were observed by HE staining.Western blot was applied to detect the expressions of Nrf2 and keap 1 proteins in liver tissues.Results:Compared with the control group,the levels of MDA in liver homogenate of Con A model mice was increased,while SOD activity was decreased.In addition,the serum levels of ALT,AST,NO,TNF-α,IFN-γ,IL-4 and IL-6 were significantly increased.15 g/kg mDSS decreased the MDA level of liver homogenate and increased SOD level significantly.5 and 15 g/kg mDSS inhibited the elevation of ALT,AST,NO,TNF-α,IFN-γ,IL-4 and IL-6 levels in serum,and up regulated the expressions of Nrf2 and Keap1 protein.Conclusion:The modified Danggui-Shaoyao-San has the obvious protective effects on Con A-induced acute liver injury in mice.Its mechanism may be related to regulation of Nrf2 signaling pathway.
