摘要 目的研究双歧杆菌脂磷壁酸(LTA)对刀豆蛋白A(Con A)活化小鼠肝细胞肿瘤坏死因子-α(TNF-α)和一氧化氮(NO)水平的影响,探讨临床应用LTA治疗免疫性肝损伤的可能性。方法将用胶原酶灌注法获取的小鼠肝细胞分为对照组(达尔伯克改良伊格尔培养基完全培养液)、LTA+Con A处理组(LTA终浓度分别为1.0、2.0、4.0μg.L-1,Con A终浓度为10 mg.L-1)和Con A组(Con A终浓度为10mg.L-1);细胞处理24 h后,采用酶联免疫吸附法测定肝细胞上清液中TNF-α和NO水平,反转录-聚合酶链反应检测肝细胞TNF-αmRNA水平。结果 ConA组肝细胞上清液中TNF-α和NO水平及肝细胞TNF-αmRNA水平均显著高于对照组(P<0.05),不同浓度的LTA处理后上述各指标均较Con A组显著降低(P<0.05),且LTA的作用与其浓度呈一定的梯度关系。结论 LTA可能通过抑制Con A活化的小鼠肝细胞体外TNF-α和NO的生成发挥对免疫性肝损伤的保护作用。 Objective To observe the effect of lipoteichoicacid (LTA) extracted from Bacillus bifidus on the levels of tumor necrosis factor-a(TNF-a) and nitric oxide (NO) in hepatocytes of mince stimulated by concanavalin A (Con A) in vitro ,in order to explore the clinical application of LTA in immunological hepatic injury. Methods The hepatocytes isolated from mice by collagenase perfusion method were divided into control group (overall Dulbecco modified Eagle medium culture solution ), LTA + Con A group ( LTA final concentration was 1.0, 2.0 and 4.0 μg · L ^-1, respectively ; Con A final concentra- tion was 10 mg . L^-1 )and Con A group( Con A final concentration was 10 mg · L^-1 ). After the hepatocytes cultured for 24 hours, the levels of TNF-a and NO in hepatocyte supernatant were detected by enzyme linked immunosorbent assay, TNF-a mRNA level of hepatocytes was detected by reverse transcription-polymerase chain reaction. Results The TNF-a and NO lev- els in hepatocyte snpernatant and TNF-a mRNA level in hepatocytes of Con A group were significantly higher than those of control group ( P 〈 0.05 ) ; after the hepatocytes were treated with different concentrations of LTA, the levels of TNF-a, NO and TNF-a mRNA were significantly decreased(P 〈 0.05) ;and the effect of LTA was correlated to its concentrations. Conclusion LTA may have protective effect on immunological hepatic injury by inhibiting the production of TNF-a and NO in hepato- cytes of mice stimulated by Con A in vitro.
