摘要
目的 将能与细胞表面整合素受体特异性结合的多肽共价结合于改性聚 (丙交酯 co 乙交酯 ) (PLGA)材料上 ,以提高材料对骨组织工程种子细胞的黏附能力。方法 用固相合成法合成 [K] 16 GRGDSPC多肽并鉴定 ;取 1g/L多肽溶液与已用交联剂预处理的改性PLGA材料室温下反应 ,用X射线光电子光谱法 (XPS)表征。结果 PLGA改性后引入了含活性基团的氨基酸序列 ,所合成的多肽序列为 [K] 16 GRGDSPC ,纯度达 95 %以上 ;XPS检测表明多肽成功地固定于PLGA多孔基质材料上。结论 在PLGA基质材料上固定 [K] 16 GRGDSPC多肽 ,为实现对骨组织工程种子细胞的高效黏附奠定了基础。
ObjectiveTo improve the adhesive ability of the seed cells to the scaffold materials in bone tissue engineering through the way of integrating the polypeptides,which can specifically bind to the cell membrane integrin receptor,with the modified poly (lactic acid-co-glycolic acid) (PLGA) by covalent bonds.MethodsThe _ 16-GRGDSPC peptides which contained -RGD-sequences were synthesized using solid phase chemistry and then purified.The materials were coupled with 1 g/L polypeptides via the heterobifunctional reagent at room tempreture for 12 h.The polypeptides modification was characterized by using X-ray photoelectron spectroscopy (XPS).ResultsBio-active amino acid sequences were introduced in the modified PLGA- copolymer.The results of HPLC and Mass spectrograph have proved that the polypeptide synthesized was _ 16-GRGDSPC with a purity of over 95%.XPS revealed that the polypeptide of _ 16-GRGDSPC could successfully be bond to the scaffold material of PLGA.ConclusionIntegrating the polypeptide of _ 16-GRGDSPC with the scaffold of PLGA is of fundamental importance to more effective binding of seed cells in the bone tissue engineering.
