摘要
目的:评价5种不同摩尔比的聚丙交酯/乙交酯(PLGA)胆道支架在体外人胆汁中的降解规律及其径向支撑力的变化规律.方法:取5种比例(90L/10G,80L/20G,70L/30G.60L/40G.50L/50G)的PLGA支架各32个,每一比例分成8组,分别浸泡于装有新鲜人胆汁的试管中并置于37%恒温震荡器.每周观察支架的大体形态,测量其质量和径向支撑力。扫描电镜观察支架表面形态.结果:①支架的降解时间随丙交酯含量的减少而缩短,其降解高峰依次为35~42d,28—35d,21—28d,14~18d和8—15d;②支架的径向支撑力随丙交酯含量的减少而降低,其能够在胆道中维持支撑的时间分别为5,4,3,2和1wk.结论:PLGA材料具有可调节的生物降解性和良好的力学特性,适宜制成胆道支架.
AIM: To evaluate the biodegradation law and the change in radical force of 5 different mole-ratios of biliary stents made of poly (lactic-co-glycolic acid) (PLGA) in human bile in vitro. METHODS: Thirty-two PLGA biliary stents of 5 different mole-ratios (90L/10G, 80L/20G, 70L/30G, 60L/40G, 50L/ 50G) were respectively divided into 8 groups. All of them were independently submerged into shaking test tubes with fresh human bile and incubated at 37℃. Their shapes were observed and their weights and radical forces were measured every week. The surfaces were observed by scanning electron microscope. RESULTS: (1) With the content of PLA decreasing, the degradation time of the stent became shorter. The degradation peak times of 5 different mass-ratios of biliary stents were 35 - 42 d, 28 - 35 d, 21 -28 d, 14 - 18 d and 8 - 15 d, respectively; (2) With the content of PLA decreasing, the radical force of the stent became weaker. The times for which 5 different mass-ratios of biliary stents could sustain the support in the bile duct were 5, 4, 3, 2 and 1 wk, respectively. CONCLUSION: PLGA material has adjustable biodegradation and good mechanical properties, so it is suitable for preparation of biliary stent.
