摘要
目的 :检测妊高征胎盘的Syndecan - 1表达水平 ,探讨妊高征的发病机制。方法 采用免疫组织化学法结合计算机辅助图像分析技术 ,定量分析 10例正常妊娠组与 30例妊高征组Syndecan - 1在胎盘合体滋养层细胞的表达。结果 10 0 %正常妊娠组胎盘合体滋养层细胞免疫组织化学染色呈极强阳性 (+++)或强阳性 (++)或阳性 (+)表达 ;妊高征组胎盘合体滋养层细胞免疫组织化学染色呈弱阳性 (± )或无表达 (- )的比例和呈强阳性或阳性表达的比例分别为 5 3.33%和 4 6 .6 7%。计算机图像分析正常妊娠组和轻度、中度、重度妊高征组的平均光度值分别为 0 .4 74± 0 .343和 0 .332± 0 .10 9、0 .35 3± 0 .0 9、0 .35 0± 0 .0 6 8;统计分析表明中度妊高征与重度妊高征组的平均光度值差异无显著性意义 (P >0 .0 5 ) ,正常妊娠组与轻、中、重妊高征组组间的平均光度值差异均有显著性意义 (P <0 .0 5 ) ,轻度妊高征组与中、重度妊高征组组间的平均光度值差异均有显著性意义 (P <0 .0 5 )。结论 胎盘合体滋养层细胞Syndecan - 1蛋白表达减少可能是妊高征的发病机制之一。
Objective: To investigate the expression of Syndecan-1 on placental trophoblasts of pregnancy induced hypertension(PIH) so as to explore the pathogenesis of PIH. Methods: Inmmunohistochemistry and computer assisted image processing analysis system were used to detect the expression quantitative evaluation of Syndecan-1 on placenta syncytiotrophoblast in 10 normal pregnancy (control groups) and 30 patients with PIH. Results: In 100% of normal placentas the immunoreactivity with antibody CD138 exhibited very intense staining or intense staining or moderate staining, whereas 53.33% of PIH placentas was negative or week staining and 46.67% of PIH placentas was intense staining or moderate staining. Computer assisted image processing analysis was carried out on average brightness Syndecan-1 staining value of normal placentas and the different degrees (mild, moderate and severe ) of PIH which were 0.474±0.343?0.332±0.109?0.353±0.09 and 0.350±0.068 respectively. There was not difference statistic significantly between the moderate PIH group and the severe PIH group( P >0.05)but statistic significantly between control group and PIH groups( P <0.05).There was statistic significantly between mild PIH group and moderate and severe PIH grorps ( P <0.05). Conclusion: It shows that the reduced expression of Syndecan-1 in placenta syncytiotrophoblast of PIH may be one of the important factors responsible for the pathogenesis of PIH.
