摘要
目的观察皮质醇(Cor)对离体胎盘组织分泌促子宫收缩激素的影响。方法取健康妇女妊娠8-10周的人流胚胎组织中绒毛膜和蜕膜以及健康自然分娩初产妇的胎盘羊膜,在妊娠末期母体雌二醇和孕酮血浆浓度环境下体外培养。当处于指数生长期时,分别给予妊娠末期母体血浆浓度的0倍(对照组)、1倍(2ng/ml)、3倍(6ng/ml)和6倍(12ng/ml)的Cor刺激。每个浓度组各设10个复孔,培养4h后取上清液,测定催产素(OT)、促肾上腺皮质激素释放激素(CRH)、前列腺素E2(PGE2)和前列腺素F2α(PGF2α)浓度。结果绒毛膜组织分泌CRH和PGE2、蜕膜组织分泌PGE2和PGF2α和羊膜组织分泌PGE2的量,三个干预组均较对照组呈浓度依赖性增多(P<0.05或P<0.01)。结论 Cor可呈浓度依赖性促进体外培养的绒毛膜组织分泌CRH和PGE2、蜕膜组织分泌PGE2和PGF2α和羊膜组织分泌PGE2。
Objective To investigate the effects of cortisol on the secretion of promoting uterine contraction hormones in placenta tissues in vitro. Methods The chorion and decidua of 8-10 weeks old embryo from healthy women underwent induced abortion and the amniotic membrane of placenta from spontaneous labor healthy primiparas were cultured in vitro with human terminal pregnancy maternal plasma concentrations of estradiol and progesterone. When in exponential phase, the tissues cultured were stimulated by a solution without cortisol(group 0) or with one time(group A, 2 ng/ml), three times(group B, 6 ng/ml) or six times(group C, 12 ng/ml) of terminal pregnancy maternal plasma concentration of cortisol,which were put in ten holes for each concentration group and cultured for 4 hours. Then the supernatant was collected for the measurements of oxytocin(OT), adrenocorticotropic hormone releasing hormone (CRH),prostaglandin E2 (PGF_e) and prostaglandin F2a (PGF2a). Results The secretions of CRH and PGEe by chorion tissue, PGE2 and PGF2o by decidual tissue, and PGE2 amniotic membrane tissue were in an order of group C 〉group B 〉group A, which were all more than those in group 0 in a concentration-dependent manner(P〈0. 05 or P〈0. 01). Conclusion Cortisol can promote chorionic tissue to secrete CRH and PGE2, decidual tissue to secrete PGE2 and PGF2a, and amniotic membrane to secrete PGE2 in vitro in a concentration-dependent manner.
