摘要
目的 观察内皮素 - 1(ET 1)对心脏心肌细胞和成纤维细胞DNA和蛋白合成作用。方法 采用胰酶消化法培养新生Sprague Dawley(SD)大鼠心脏心肌细胞和成纤维细胞 ,以3 H 胸腺嘧啶核苷 (3 H TdR)、3 H 亮氨酸 (3 H Leu)掺入法测定两种细胞DNA、蛋白合成 ,Bradford法测两种细胞总蛋白含量 ,逆转录聚合酶链反应法 (RT PCR)法测心肌细胞心钠素 (ANP)mRNA的表达。结果 对于心肌细胞 ,ET 1(10 -9~ 10 -7mol/L)显著促进3 H TdR掺入率、较对照组分别增加 0 31± 0 0 79(P <0 0 1)、1 193± 0 2 6 7(P <0 0 0 1)、1 336± 0 2 78(P <0 0 0 1) ,3 H Leu掺入率增加 0 2 2 3± 0 0 5 8(P <0 0 1)、1 0 5 4± 0 2 0 2 (P <0 0 0 1)、1 0 74± 0 2 0 4 (P <0 0 0 1) ,呈剂量依赖性 ,ANPmRNA表达明显增加 ;对于成纤维细胞 ,ET 1(10 -8mol/L)促进3 H TdR掺入率 ,较对照组增加 0 2 4 8± 0 0 2 (P <0 0 5 )、3 H Leu掺入率较对照组增加 0 4 34± 0 0 4 1(P <0 0 1)。结论 ET 1促进心肌细胞肥大和心脏成纤维细胞增殖 ,且对心肌细胞的作用强于对成纤维细胞的作用。提示ET 1促心肌肥厚主要使促进心肌细胞肥大 ,对间质纤维化作用较弱。
Objective To investigate the effect of endothelin-1 on protein and DNA synthesis in cultured cardiomyocytes and fibroblasts. Methods The cells were isolated by trypsin digestion method. DNA and protein synthesis were measured by 3H-TdR and 3H-Leu incorporation and protein content was measured by Bradford method. ANP mRNA expression of cardiomyocyte was determined by RT-PCR. Results In cardiomyocytes, ET-1(10 -9 -10 -7 mol/L) significantly increased the 3H-TdR and 3H-Leu incorporation in a dose-dependent manner. Additionally, ET-110 -8 mol/L promoted the ANP mRNA expression in cardiomyocyte. In fibroblasts, ET-110 -8 mol/Lsignificantly increased the 3H-TdR and 3H-Leu incorporation rate. Conclusion ET-1 induces hypertrophy in cardiomyocytes and the proliferation for cardiac fibroblasts.
