摘要
将新鲜猪小肠超声破碎 ,经高温处理、乙酸浸提后 ,再经SephadexG10 0和SephadexG2 5凝胶柱过滤层析 ,收集各组分并用琼脂糖弥散法检测各组分的抗菌活性。收集具有抗菌活性的组分 ,经TricineSDS—PAGE ,考马斯亮蓝染色后分析为一条蛋白带 ,分子量约为 5 972Da左右 ,得到电泳纯的猪小肠抗菌肽。该抗菌肽对大肠杆菌C84 0 0 8株、金黄色葡萄球菌ATCC2 5 92 3株、绿脓杆菌ATCC2 785 3株均表现出较强的抗菌活性 ;
After the small intestines of pig were treated by ultrasonic treatment, heating and 5% acetic acid extraction, antimicrobial peptides were isolated from the intestine of pig by Sephadex G100 and Sephadex G25 gel filtration chromatography. The samples were analyzed by Tricine SDS polyacrylamide gel electrophoresis. The results showed that an antimicrobial peptide was purified and its molecule weight was about 5 972 Da. The antimicrobial peptide, named PSIP, strongly inhibited Escherichia coli ATCC84008, Staphylococcus aureus ATCC25923 and Pseudomonas aeruginosa ATCC27853, and neutralized chicken infectious bronchitis virus H120.
