摘要
将新鲜草鱼肠道组织经超声破碎、乙酸浸提后,再经Sephadex G50和Sephadex G25凝胶柱过滤层析,收集各组分,采用药敏纸片法检测各组分的抗菌活性.收集具有抗菌活性的组分,经Tricine SDS-PAGE,考马斯亮蓝染色后分析获得1条较明显的蛋白带,相对分子质量约为6 200.该抗菌肽对大肠杆菌、金黄色葡萄球菌和嗜水气单胞菌均表现出较强的抗菌活性,说明抗菌肽对维持鱼类肠道微生物区系稳定具有重要作用.
After the small intestines of grass carp being treated by ultrasonic treatment and a 5% acetic acid extraction, antimicrobial peptides were isolated from the intestine of grass carp by Sephadex G50 and Sephadex G25 gel filtration chromatography. The samples were analyzed by Tricine SDS polyacrylamide gel electrophoresis. The results showed that an antimicrobial peptide was purified and its molecule weight was about 6 200. The antimicrobial peptide strongly inhibited Escherichia coli, Staphylococcus aureus and Aeromonas hydrophila. It suggested that the antimicrobial peptide of fish intestine is playing an important role in maintenance to the stability of intestinal microbial flora.
