摘要 用硫酸铵盐析法从抗猪瘟血清中提取免疫球蛋白 ,应用搅拌法标记荧光素 ,硫酸铵盐析、SephadexG 2 5层析和肝粉吸收三法结合去除标记物之游离荧光素 ,制备猪瘟荧光抗体。经测定 ,标记的荧光抗体工作稀释度为 1∶16~ 1∶32。用标记的荧光抗体诊断猪瘟 ,从 39份可疑病料中检出阳性病例 12份。 The methods of preparation and application of fluorescent antibody against classical swine fever (CSF) were introduced .Immunoglobulin was extracted from swine serum against CSF by means of using (NH_4)_(2)SO_4 fractionation, and was labelled with fluorescein isothiocyanate (FITC) using agitation methods. Unbound FITC was removed by (NH_4)_(2)SO_4 fractionation, Sephadex G-25 column and liver powder absorption. The working dilution of fluorescent antibody was 1∶16-1∶32.Using fluorescent conjugated antibody to diagnose CSF, 12 positive examples were found from 39 suspicious examples.
