苦参碱诱导K562细胞分化方向的研究 被引量:13

摘要 [目的]体外研究苦参碱诱导白血病细胞株K562的分化方向。[方法]0.2mg/ml浓度苦参碱诱导K562细胞7天,观察苦参碱作用前后K562细胞形态学、细胞化学及细胞谱系特征性标记变化。[结果]K562细胞出现分化的形态特征,联苯胺染色及过碘酸—希夫反应(PAS)阳性细胞数分别由5%上升至31%,3%上升至35%,α鄄醋酸萘酚酯酶(α鄄NSE)、中性粒细胞碱性磷酸酶(NAP)阳性细胞数分别由36%下降至3%,22%下降至0,CD34、CD41表达阳性细胞数分别由1.1%上调至6.4%,23.8%上调至69.2%,CD68表达阳性细胞数由8.6%下调至1.1%,CD3、CD45RO、CD56及CD20表达阳性细胞数无明显变化。[结论]苦参碱是白血病细胞K562良好诱导分化剂,可诱导K562细胞向髓系非单核巨噬细胞方向分化。 The potential differentiating pathway of leukaemia cells K562induced by matrine in vit-ro was studied.Differentiation induction experiment was performed by culturing K562cells with the presence of0.2mg/ml matrine for7d.The changes of morphological,cytochemical and cytophyletic char-acteristic marker of K562cells were observed before and after the treatment with matrine.Differen-tiating morphologic features appeared with the increase of benzidine-positive cells from5%to31%and PAS-positive cells from3%to35%.The NSE-positive cells decreased36%to3%and completely.All ALP-posi-tive cells disappeared22%to0.CD34and CD41expression was up-regulated,CD68expression was down-regulated.The expression of CD3,CD45RO,CD56and CD20did not changed significantly.[Conclusion]Matrine may become the high potency inducer in treatment of leukaemia and it can induce K562differentia-tion along a nonmonocytic series myelocyte pathway.
