苦参碱逆转人白血病K562/ADM细胞对阿霉素耐药性的研究 被引量:13

摘要 [目的 ]探讨苦参碱对人白血病K5 6 2 /ADM细胞对阿霉素耐药性的逆转作用。 [方法 ]MTT法测定苦参碱的细胞毒性及其对K5 6 2 /ADM细胞药敏性的影响 ,荧光分光光度法检测细胞内药物浓度的改变 ,流式细胞术检测耐药细胞凋亡百分率的变化。 [结果 ]苦参碱的非细胞毒性剂量为 5 0μg/mL ,低细胞毒性剂量为 12 5 μg/mL。 5 0 μg/mL苦参碱可增加K5 6 2 /ADM细胞内阿霉素(ADM )浓度和K5 6 2 /ADM细胞凋亡百分率 ,使K5 6 2 /ADM细胞的IC50 由原来的 35 .2 μg/mL降低至 15 .8μg/mL ,其逆转倍数为 2 .2倍。[结论 ]苦参碱可部分逆转人白血病K5 6 2 /ADM细胞对阿霉素的耐药性 ,其逆转机制与增加细胞内药物积累有关。 [Objective] To investigate the reversal effect of adriamycin (ADM) resistance by matrine in leukemia multidrug resistance cell line K562/ADM. [Methods] Cytotoxicity of matrine and the sensibility to adriamycin (ADM) of K562/ADM cell line, leukemia cell line with multidrug resistance, were studied through MTT assay. The concentration of intracellular ADM was detected by spectrofluorometry. Cell apoptosis percentage was detected by flow cytometry . [Results] The non-cytotoxic dose of matrine was 50 μg/mL and the low cytotoxic dose was 125 μg/mL to K562/ADM cell line. The matrine in 50 μg/mL could significantly increase the intracellular accumulation of ADM in K562/ADM cell line and apoptosis percentage in K562/ADM. The medium inhibition concentration (IC 50 ) was obviously reduced from 35.2 μg/mL to 15.8 μg/mL, with a reversal ratio of 2.2 as compared to its parental cell line. [Conclusion] Matrine is able to enhance the cytotoxicity of ADM and partly reverse the ADM resistance of K562/ADM cell line, through increasing accumulation of intracellular ADM.
