小叶榕对过敏性哮喘豚鼠模型的药效作用及其机制研究 被引量:3

摘要 目的:探究小叶榕对过敏性哮喘豚鼠模型的药效作用及其机制,为后续的药理机制和药效物质基础研究提供实验依据。方法:将豚鼠随机分为空白对照组、模型对照组、阳性对照组(孟鲁司特钠)、小叶榕干浸膏低剂量组、小叶榕干浸膏中剂量组、小叶榕干浸膏高剂量组,每组10只。除空白对照组外,其余5组均通过腹腔注射卵蛋白致敏和吸入卵蛋白诱导哮喘发生,建立豚鼠过敏性哮喘模型。观察各组豚鼠引喘潜伏期、咳嗽次数、肺组织病理学、肺泡灌洗液(BALF)中白细胞总数和嗜酸粒细胞(EOS)百分率、血清中白细胞介素5(IL-5)、白细胞介素4(IL-4)、血清免疫球蛋白E(Ig E)含量的改变。结果:与空白对照组相比,模型组豚鼠肺组织有明显病理改变,BALF中白细胞总数和EOS百分率、血清中细胞因子IL-5、IL-4、Ig E含量均增加(P<0.05)。各治疗组与模型组对比,引喘潜伏期均增加,咳嗽次数减少,BALF中白细胞总数和EOS百分率、血清中IL-5、IL-4、Ig E含量明显降低(P<0.05)。小叶榕低、中剂量组与阳性对照组对比,咳嗽次数增加,BALF中白细胞总数和EOS百分率、血清中IL-5、IL-4、Ig E含量明显增加(P<0.05)。结论:小叶榕能够改善过敏性哮喘豚鼠的哮喘症状、减轻肺组织病变情况。其作用机制可能与调节参与炎症反应的细胞因子IL-4、IL-5,血清中Ig E和减少EOS浸润有关。 Objective:To explore the pharmacodynamic effect and underlying mechanism of Ficusmicrocarpa in guinea pigs with allergic asthma to provide experimental basis for the follow-up study on the pharmacological mechanism and pharmacodynamic material basis of Ficusmicrocarpa.Methods:Totally 60 guinea pigs were randomly divided into six groups:the normal group,the model group,the positive group(montelukast sodium),low-dose group of Ficusmicrocarpa extract,middle-dose group of Ficusmicrocarpa extract and high-dose group of Ficusmicrocarpa extract with 10 guinea pigs in each.Except for the normal group,the experimental allergic asthma model of guinea pigs was established by intraperitoneal injection of ovalbumin and inhalation of ovalbumin.The latency of asthma induction,cough frequency,lung histopathology,total white blood cells and EOS percentage in BALF,and serum levels of IL-5,IL-4 and Ig E were observed.Results:Compared with that in the blank control group,the lung tissue in the model group had obvious pathological changes.The total number of white blood cells,the percentage of EOS and the serum levels of cytokines IL-5,IL-4 and Ig E in BALF increased(P<0.05).Compared with the model group,the treatment group increased the incubation period and the times of cough decreased.The total number of leukocytes and EOS in BALF,serum IL-5,IL-4 and Ig E levels were significantly decreased(P<0.05).Compared with that in the positive control group,the times of cough increased,the total number of white blood cells and EOS in BALF,and the levels of IL-5,IL-4 and Ig E in serum increased significantly(P<0.05).Conclusion:Ficusmicrocarpa can improve asthma symptoms and alleviate lung pathological changes in allergic asthmatic guinea pigs.The therapeutic mechanism may be related to the regulation of cytokines IL-4,IL-5 and serum total Ig E,and the reduction of EOS infiltration.
