创伤性股骨头坏死血清差异蛋白质组学的研究 被引量:8

A study on differential proteomics in traumatic osteonecrosis of the femoral head

摘要 [目的]探讨创伤性股骨头坏死(TONFH)的血清蛋白质组学差异质,寻找并鉴定TONFH的潜在血清生物学标志物。[方法]分别采集TONFH患者(实验组)、正常体检者(对照组)血清各5例,采用TMT联合2DLC-MS/MS技术进行蛋白组学分析,寻找并鉴定差异表达的蛋白质。[结果]质谱共筛选出有意义差异蛋白344个,并鉴定出15种显著性差异蛋白(P<0.05),其中与正常组对比,实验组中抗粘蛋白1轻链可变区、补体C5(C5)、冷凝集蛋白FS-2 H链、纤维连接蛋白(FN1)、妊娠区带蛋白(PZP)、β肌动蛋白(ACTB)、甘露糖结合蛋白C(MBL2)等7种蛋白表达上调,Igλ链V区4A、多巴胺β-羟化酶(DBH)、免疫球蛋白重链可变区、CD5抗原(CD5L)、免疫球蛋白J链(JCHAIN)、补充C1r子组件(C1R)、抗凝血酶III (Antithrombin-III)、凝溶胶蛋白(GSN)等8种蛋白表达下调。[结论]采用同位素相对标记与绝对定量技术(TMT)联合二维液相色谱-串联质谱(2D-LC-MS/MS),成功筛选出了TONFH的血清差异表达蛋白质,为寻找TONFH特异性标志物及进一步阐明TONFH的发病机制提供了依据。 [Objective] To investigate the serum proteomics of traumatic osteonecrosis of the femoral head(TONFH) and identify potential serum biomarkers of TONFH. [Methods] Serum samples were obtained from 5 patients with TONFH and 5 normal persons. The tandem mass tags(TMT) combined with 2 D-LC-MS/MS techniques were used for proteomic analysis to find and identify differentially expressed proteins. [Results] A total of 344 genes with significant differences were screened out by mass spectrometry, and 15 proteins with significant differences were identified(P<0.05). Compared with the normal group, 7 proteins proved up-regulated in the TONFH group, including the anti-viscoprotein 1 light chain variable region, complement C5(C5), condensing protein FS-2 H chain, fibronectin(FN1), pregnancy zone band protein(PZP), β actin(ACTB), and mannose-binding protein C(MBL2). Conversely, 8 proteins was down-regulated in the TONFH group, such as Igλ chain V region4 A, dopamine β-hydroxylase(DBH), immunoglobulin heavy chain variable region, CD5 antigen(CD5 L), immunoglobulin J chain(JCHAIN), complementary C1 r subcomponent(C1 R), and antithrombin-III(Gelsolin, GSN). [Conclusion] The differentially expressed proteins of TONFH were successfully screened by isotope relative labeling and absolute quantification(TMT)combined with two-dimensional liquid chromatography-tandem mass spectrometry(2 D-LC-MS/MS) for the purpose of searching for TONFH specific markers, which should provide the basis for further elucidation of the pathogenesis of TONFH.
