摘要
目的评价血管生成抑制剂 TNP-470与碘化油混合栓塞对兔肝癌的抑瘤效果.方法将 VX_2瘤细胞接种于24只新西兰白兔肝左叶,建立兔肝癌模型,随机分4组,每组6只.利用导管经肝动脉分别给碘油、TNP-470,TNP-470+碘油及生理盐水于不同组,1 wk 后观察各组肿瘤体积、肿瘤微血管密度及血清 AST 水平,并观察存活期及组织病理切片.结果 TNP-470+碘油组生长率为(0.62±0.28)与对照组(3.48±1.17)相比差异有显著性(P<0.01),与栓塞组(1.28±0.23)、TNP-470组(1.18±0.26)相比差异有显著性(P<0.05).混合栓塞组微血管密度(MVD 值)(9.98±5.71)与对照组(23.12±9.67)相比差异有显著性(P<0.01).混合栓塞组存活期(53.0±2.0)与对照组(40.5±3.0)相比差异有显著性(P<0.05).混合栓塞组血清 AST 水平治疗前后变化与其他处理相比差异无显著性(P>0.05),与对照组相比差异有显著性(P<0.05).混合栓塞组病理 HE 染色切片示:有早期坏死表现.结论 TNP-470与碘油混合栓塞组可大大降低肿瘤生长率,减小微血管密度,延长存活期,并对肝功损害是可逆的,其抑瘤效果更强.
AIM To evaluate the anti-tumor effects of angiogenesis inhibitor TNP-470 combined with lipiodol which was injected and embolized via hepatic artery of liver cancer of rabbits. METHODS Vx_2 carcinoma cells were surgically implanted into the left liver lobe of 24 male New Zealand white rabbits,which were divided into 4 groups at random,with 6 rabbits in each group.Lipiodol,TNP-470,lipiodol plus TNP-470 and physiological saline were injected in different groups via hepatic artery to embolize liver cancer of rabbits.One week later,the volume of tumor, microvessel density,the serum level of AST,the survival period and pathological slices were observed. RESULTS In the group of TNP-470 plus lipiodol,the tumor growth rate (0.62±0.28) was significantly lower than the other treated groups (1.28±0.23,1.18±0.26) (P<0.05),being significantly lower than the untreated control group (3.48±1.17) (P<0.01).The value of MVD in TNP-470 plus lipiodol group (9.98±5.71) was less than the untreated control group (23.12±9.67,P<0.01).The survival period of the former group (53.0±2.0) was significantly higher than that of the untreated control group (40.5±3.0,P<0.05),The serum level of AST of TNP-470 plus lipiodol group was not higher than the other treated groups (P>0.05),but significantly higher than the untreated control group (P<0.05).The H.E.stained pathological tissue showed some signs of inchoate necrosis. CONCLUSION TNP-470 plus lipiodol greatly decreases the tumor growth rate,and reduces the value of microvessel density,and prolongs the survival period.It has a stronger inhibitory effect than other groups.
