摘要
目的 :观察柔红霉素 (DNR)和bax基因转染联合应用对兔实验性增生性玻璃体视网膜病变 (proliferativevitre oretinopathy ,PVR)形成的影响 .方法 :将 2 .5× 10 5个正常视网膜色素上皮 (retinalpigmentepithelial,RPE)细胞和 3.5×10 5个经bax基因转染的RPE细胞注入兔玻璃体腔 ,观察诱发兔眼PVR形成的能力 ,同时观察 10nmolDNR对PVR形成的抑制作用 .结果 :在 3,7,14 ,2 1和 2 8d时RPE组PVR的平均分级为 :0 .6 2 5 ,2 .0 0 0 ,3.0 0 0 ,4 .12 5和 4 .2 5 0 ,基因转染细胞组PVR的平均分级为 :0 .5 0 0 ,0 .875 ,2 .0 0 0 ,3.12 5和3.6 2 5 ,后者PVR分级均低于RPE细胞组 (P <0 .0 5 ) .2 8d时正常细胞组视网膜脱离发生率为 10 0 % (n =8) ,bax基因转染细胞组的视网膜脱离发生率为 6 2 .5 % (n =8) .在 7,14 ,2 1和 2 8d时 ,DNR对正常细胞组PVR的抑制率分别为6 0 % ,6 4 % ,5 7%和 5 7% ,对基因转染细胞组PVR分级的抑制率分别为 73% ,71% ,6 8%和 6 9% ,后者普遍大于前者 .结论 :DNR可以抑制PVR的形成 。
AIM: To observe the combined effects of daunorubicin and bax gene transfection on the formation of rabbit proliferative vitreoretinopathy(PVR). METHODS: 2.5 ×10 5 retinal pigment epithelial (RPE) cells and 3.5×10 5 bax gene transferred RPE cells were injected into the vitreous cavity and the difference was observed between the development of PVR and the inhibitory effects of 10nmol daunorubicin on PVR formation. RESULTS: The mean PVR stage of normal RPE group was 0.625,2.000,3.000,4.125 and 4.250 on 3, 7, 14, 21 and 28 d respectively, while that in the bax gene transferred RPE cell group was 0.500,0.875,2.000,3.125 and 3.625. The mean values of bax gene transferred RPE cell group were lower than those of RPE cell group( P <0.05). By 28 days, all the 8 eyes of normal RPE cell group developed traction retinal detachment (TRD), while in the bax gene transferred RPE cell group, only 62.5% eyes developed TRD. The inhibitory rates of daunorubicin on PVR grades of the RPE group were 60%, 64%, 57% and 57% on postoperative day 7, 14, 21 and 28, respectively, and those of the bax gene transferred RPE cell group were 73%, 71%, 68% and 69% respectively. The inhibitory rates were higher than those of the normal RPE group ( P <0.01) at all time points. CONCLUSION: Daunorubicin can inhibit the formation of experimental PVR of rabbits and bax gene transfection can enhance the prevention efficiency.
