摘要
目的:探讨创伤后应激障碍(posttraumaticstressdisorder,PTSD)样精神与行为异常的病理生理基础,为其治疗途径提供新思路。方法:将72只雄性Wistar大鼠随机分组为海马阈下电刺激组(SE,n=32)、海马电极埋植对照组(CE,n=32)和正常对照组(NC,n=8),利用频率25Hz、波宽1ms、串长10s、串隔7min、强度100μA的恒流、单脉冲电流,反复惊厥阈下电刺激海马,并定量检测了实验动物海马组织匀浆及线粒体Na+-K+-ATP酶、Ca2+-ATP酶活性改变。结果:电刺激停止后12h阈下刺激大鼠海马细胞线粒体Na+-K+-ATP酶活性即明显下降为(0.56±0.15)mmol/(kg·s),(F=4.348,P<0.01),48h为(0.61±0.17)mmol/(kg·s),(P<0.05)仍显著低于NC组(0.84±0.22)mmol/(kg·s);24h线粒体Ca2+-ATP酶活性亦明显降低为(0.53±0.14)mmol/(kg·s),(F=4.999,<0.05,72h为(0.60±0.16)mmol/(kg·s)仍显著低于NC组(0.83±0.22)mmol/(kg·s)。结论:海马组织,特别是海马细胞线粒体钠钾泵与钙泵功能受损,在实验动物长时程PTSD样情感行为异常发生发展中可能有重要意义。
To discuss the pathophysiology basis of posttraumatic stress disor de r(PTSD-like)psycho and behaviour abnormity in attempt to provide a new method in treatments.METHODS:Seventy-two male Wistar rats were ra ndomly divided in to three groups:hippocampus under-threshold electric stimulation group(SE,n =32),hippocampus electrode-burying control group(CE,n=32)and normal control group(NC,n=8).Hippocampus were continuously stimulated by constant monopulse electricity,wi th 25Hz frequency,1ms wave length,10s cluster length,7min cluster interv al and 100μA strength under eclampsia threshol d.The enzymatic activity c hanges of Na + -K + -adenosine triphos-phate enzyme (ATPase)and Ca 2+ -ATPase in hippocampal homogenate o f the experimental animals and mitochon d ria were detected in quantitation.RESULTS:The enzymatic activity of Na + -K + -ATPase in hippocampus mi-tochondria decreased obviously(0.56±0.1 5)mmol /(kg·s)(F=4.348,P<0.01)in under-threshold electric s timul ation group after 12hours of electric stimulations as well as(0.61 ±0.17)mmol /(kg·s)(P<0.05)after 48hours,which were significa ntly lower than NC group(0.84±0.22)mmol /(kg·s)the enzymatic acti vity of Ca 2+ -ATPase in hippocampus mito-chondria also decreased obviously i nto(0. 53±0.14)mmol /(kg·s)(F=4.999,P<0.05)after 24hours of electric stimulations as well as(0.60±0.16)mmol /(kg·s)after 7 2hours,which were significa ntly lower than NC group(0.83±0.22)mmol /(kg·s).Functional damages of the hippocamp us,especially the Na-K-pump and Ca 2+ -pump in hippocampal mitochondria may have an im-portant significance in t he occurre nce and development of long-term PTSD-like psycho and behaviour abn ormit
