摘要
目的:建立以高效液相色谱法同时测定半夏泻心汤标准煎液中黄芩苷和盐酸小檗碱含量的方法。方法:采用Zorbax Eclipse SB-C18(250mm×4.6mm,5μm,S.N.USCL072720)色谱柱,流动相为乙腈-0.2%磷酸水溶液(加入三乙胺调pH=2.5)梯度洗脱,流速为1.0mL·min(-1),检测波长为275nm,柱温为25℃,进样量为10μL。结果:黄芩苷和盐酸小檗碱检测浓度分别在45.0～180.01μg·mL^(-1)(r=0.9992)、4.7～18.7μg·mL(-1)(r=0.999 7)浓度范围内与峰面积积分线性关系良好;精密度、稳定性、重复性的RSD均<2.0%,平均加样回收率(n=6)分别为103.33%和101.31%,RSD分别为0.74%、1.46%。结论:该方法简便、准确、重复性好,可用于同时测定半夏泻心汤标准煎液中黄芩苷和盐酸小檗碱的含量。
Objective:To establish a method for simultaneous determination of baicalin and berberine in Banxia Xiexin Decoction standard decoction by high performance liquid chromatography(HPLC).Methods:A Zorbax Eclipse SB C18(250 mm ×4.6 mm,5μm,SNUSCL072720)column was used.The mobile phase was eluted with a gradient of acetonitrile-0.2% phosphoric acid(addition of triethylamine to adjust the pH=2.5)at a flow rate of 1.0 mL/min,detection wavelength is 275 nm,column temperature is 25℃,and the injection volume is 10μL.Results:The detection concentrations of baicalin and berberine hydrochloride were linear in the range of 45.0~180.01μg/mL(r=0.9992)and 4.7~18.7μg/mL(r=0.9997),respectively.Precision stability repeatability RSD <2.0%;The average recovery rate(n=6)was 103.33% and 101.31%,respectively,and the RSD was 0.74% and 1.46%,respectively.Conclusion:This method is simple,accurate and reproducible.It can be used to simultaneously determine the content of baicalin and berberine in Banxi Xiexin Decoction standard decoction.
