摘要
目的探索维生素A对光损伤后小鼠视网膜结构和功能的保护作用。方法将12只清洁级雄性成年C57小鼠随机分为生理盐水组和维生素A组。2组小鼠于实验前行双眼视网膜电图(ERG)标准五项反应检测视网膜功能,2组小鼠分别接受1m L·kg-1生理盐水或维生素A(0.1%)腹腔内注射,连续5 d。2组小鼠按照动物光损伤模型标准制作右眼单眼光损伤模型,左眼遮盖,造模成功后继续饲养5 d,重复检测ERG。功能检测后立即取小鼠双侧眼球做视网膜组织切片,比较2组小鼠视网膜结构的变化。结果实验前2组小鼠双眼暗适应0.01ERG b波、暗适应3.0ERG a和b波、暗适应3.0震荡电位的总幅值、明适应3.0ERG b波及3.0闪烁光反应P1波总幅值的差异均无统计学意义(P均>0.05)。光损伤造模后,两组小鼠右眼与自身对照左眼相比,上述ERG 5项检测的波幅值均出现明显下降(P均<0.05);维生素A组小鼠右眼暗适应0.01ERG的b波、暗适应3.0ERG的a波和b波、暗适应3.0震荡电位的总幅值均高于生理盐水组(t=3.544,P=0.019;t=4.574,P=0.011;t=9.563,P=0.002;t=6.156,P=0.002),而明适应3.0 ERG的b波以及闪烁光反应P1波总幅值的差异无统计学意义(t=4.678,P=0.469;t=4.883,P=0.342);2组小鼠右眼的视网膜外核层(ONL)细胞层数较自身对照左眼均有所减少(P<0.05),在生理盐水组小鼠右眼的视网膜外核层(ONL)细胞层数(6.5±1.05)少于维生素A组(11.5±1.03)(t=2.301,P=0.003)。结论维生素A对光照引起的小鼠视网膜结构和功能损伤有一定的保护作用。
Objective To evaluate the effect of vitamin A in preventing retinal photic damage. Methods Twelve clean male C57 mice were randomly assigned into hydrogen-rich normal saline solution( NS) group,vitamin A group and 6 rats for each. The electroretinogram( ERG) with international standardized 5 items was recorded from all the mice before initial of experiment. NS and vitamin A of 1mL ·kg-1was intraperitoneally injected in the two groups,respectively. The ERG was repeatedly recorded after 5 days of light irradiation.The mice sacrificed at the end of the experiment and retinal sections were prepared for the histopathological examination. The functional and structural changes of retinas were compared among the different groups. Results No any significant differences were found in scotopic 0. 01 ERG b wave,scotopic 3. 0 ERG a and b waves,scotopic 3. 0 oscillatory potentials,photopic 3. 0 ERG b wave and 3. 0 flicker P1 wave in the two groups before experiment( P > 0. 05). The amplitudes of various waves of ERG were significantly declined in the right eyes compared with the left eyes after experiment among the two groups( P < 0. 05). Compared with the NS group,the mean differential values of scotopic 0. 01 ERG b wave,scotopic 3. 0 ERG a and b waves,scotopic3. 0oscillatory potentials in the right eyes of vitamin A group were significantly different( P < 0. 05),but those inphotopic 3. 0 ERG b wave were not statistically significant( P > 0. 05). The number of outer nuclear cell layers was decreased in the right eyes in these two groups in comparison with the left eyes,with a significant differences between them( P < 0. 05),and those in the vitamin A group were significantly less than NS group( P < 0. 05). Conclusion Vitamin A have a protective effect on light-induced retinal damage of C57 mice.
