摘要
目的:分析益气养阴明目汤对视网膜光损伤大鼠年龄相关性黄斑变性(ARMD)治疗作用及药理学影响。方法:选取由本院动物实验中心提供清洁剂健康SD大鼠40只,依据随机数字表法将其分成四组,对照组、益气养阴明目汤小剂量组、中剂量组及大剂量组,每组各10只,使用自制光损伤装置造模,大剂量组益气养阴明目汤灌胃剂量为3. 3ml/100g,中剂量组2. 2ml/100g灌胃,小剂量组1. 1ml/100g灌胃,对照组1. 1ml/100g生理盐水灌胃;免疫组织检测p53及Bcl-2基因阳性表达状况并检测其病理形态学,电镜察看其视网膜组织超微结构状况,并检测大鼠血清内丙二醛(MDA)、谷胱甘肽过氧化物酶(GSX-PX)及超氧化物歧化酶(SOD)含量状况。结果:中、大剂量组大鼠血清SOD、GSH-PX含量高于对照组,血清MDA含量低于对照组,差异均有统计学意义(P<0. 05);中、大剂量组大鼠p53阳性表达率低于对照组,Bcl-2阳性表达率高于对照组,差异均有统计学意义(P<0. 05)。结论:益气养阴明目汤可清除氧自由基,改善视网膜超微结构,调节Bcl-2及p53基因表达状况来改善ARMD大鼠视功能。
Objective: To analyze the therapeutic effect and pharmacological effects of Yiqi Yangyin Mingmu Decoction on macular degeneration( ARMD) in rats with photo-injury. Methods: Forty healthy Sprague-Dawley rats were provided by Animal Experimental Center of our hospital and divided into four groups according to the random number table method. Controlled group,small dose group,medium dose group and large dose of Yiqi Yangyin Mingmu Decoction. 10 rats in each group,using self-made optical damage device model,the high-dose group Yiqi Yangyin Mingmu Decoction intragastric dose of 3. 3 ml/100 g,the medium dose group 2. 2 ml/100 g gavage,the small dose group 1. 1 ml. The mice were intragastrically administrated with 100 g/100 g gavage and the controlled group was intragastrically administered with 1. 1 ml/100 g normal saline. Immunohistochemistry was performed to detect the positive expression of p53 and Bcl-2 genes and to detect the pathological morphology. Ultrastructure of the retina tissue was observed by electron microscopy and the serum of rats was examined. Content of MDA,GSX-PX and SPD. Results: The levels of serum SOD and GSH-PX in the medium and high dose groups were higher than those in the controlled group.The levels of serum MDA were lower than those in the controlled group( P〈0. 05). The rats in the medium and high dose group had p53 positivity. The expression rate was lower than that of the controlled group,and the positive rate of Bcl-2 positive expression was higher than that of the control ledgroup( P〈0. 05). Conclusion: Yiqi Yangyin Mingmu Decoction can scavenge oxygen free radicals,improve retinal superstructure,and regulate the expression of Bcl-2 and p53 genes to improve the visual function of ARMD rats.
