摘要
目的:探讨海马CA1锥体神经元对不同单色激光闪光刺激的反应,为论证激光诱发的神经系统生物学效应提供实验支持。方法:健康成年SD大鼠麻醉后,行气管插管术,开颅钻孔暴露脑面,进行在体膜片钳记录。全细胞记录稳定后,采用4种不同波长的单色激光对其眼球进行闪光刺激,记录大鼠海马神经元的反应。结果:给予大鼠闪光刺激后,其海马神经元对蓝色激光和紫色激光表现出明显的超极化反应,对红色和绿色激光刺激则没有明显的反应。蓝光引起的变化中,超极化的幅值为(8.32±1.10)m V,反应持续时间为(115.32±13.02)ms;紫光引起的超极化的幅值为(9.01±2.25)m V,刺激反应持续时间为(109.27±16.62)ms,蓝光和紫光之间没有统计学差异。50 m W、75 m W、100 m W、125 m W功率的紫色激光刺激引起的超极化幅值,分别为(7.28±0.16)m V、(9.25±0.71)m V、(10.91±0.08)m V、(12.67±0.38)m V,相关性分析显示幅值变化与刺激功率高度正相关。结论:麻醉状态下,蓝光和紫色的闪光刺激可以诱导大鼠海马神经元出现明确的抑制性反应,红光和绿光没有明显的作用。
Objective: For the purpose of providing evidences of biological effect on nervous system evoked by the laser stimulation, we investigated the response of hipocampal CA1 pyramidal neurons to the laser flash stimuli of different colors. Methods: Healthy adult Sprague Dawley rats were involved in this experiment. After anesthetized, endotraeheal incubation and craniotomy at the skull directly above the hippocampus were carried on. In vivo whole-cell patch clamp recording was conducted. During the stable recording, four kinds of flash stimuli were given to the rat eye and responses of the CA1 pyramidal neuron were recorded. Results: Blue and purple light induced obvious membrane potential hyperpo- larization in the CA1 pyramidal neurons. For blue light, the mean amplitudes of the response was 8.32 ± 1.10 mV, the average duration of the response was 115.32 ± 13.02 ms; For purple light, the mean amplitudes of the response was 9. 01±2.25 mV and the average durations of the response was 109.27 ±16.62 ms. There is no significant differences between these two groups in both amplitudes and durations of the response ( P 〉 0.05 ). Though, the red and green light didn't make any change on the membrane potential. A graded power of purple laser (50 mW,75 mW,100 mW, 125 mW) induced the gradually increased amplitudes of membrane potential hyperpolarization (7.28 ± 0.16 mV,9.25 ± 0.71 mV, 10.91 ±0.08 inV,12.67 ±0.38 mV), suggesting a high correlation. Conclusion: Blue and purple laser light can cause significant inhibitory responses in the CA1 pyramidal neurons, in comparison, there is no significant effect evoked by red and green laser stimuli.
