摘要
通过单因素的改变对核质互作型水稻线粒体DNA的RAPD反应条件进行优化 ,并在最佳条件下用OPERON公司OPA~OPM系列共 2 6 0个随机引物筛选水稻Ⅱ 32不育系、保持系、杂种一代线粒体DNA的差异片段 ,结果共获得 2 2个差异带。其中OPH19 180 0、OPJ0 9 4 0 0、OPJ18 14 0 0、OPJ18 10 0 0、OPJ2 0 30 0这几个特异片段可能与水稻雄性不育相关。我们正在用分子检测确定这些标记与细胞质雄性不育的关系。
The reaction conditions of RAPD for mitochondrial DNA of nucleo-cytoplasmic interaction rice were optimized by changing a single factor. Under the optimum conditions, Ⅱ-32 male sterility line, maintainer line and F 1 hybrid mitochondrial DNA were screened by 260 RAPD primers from OPA to OPM. As a result, 22 differential fragments were found. Among them, OPH19-1800, OPJ09-400, OPJ18-1400, OPJ18-1000 and OPJ20-300 might have relation with rice male sterility. For further study, the relationship between the markers and cytoplasmic male sterility would be studied.
