谷氨酸高产菌FTN9108的细胞融合育种及其发酵条件的研究 被引量:5

Breeding of High Glutamate-Producing Strain FTN 9108 Using the Protoplast Fusion Technique and Study on It's Fermentation Conditions

摘要 以谷氨酸生产菌天津短杆菌(Brevibacterium tianjinese)T_(6-13)为出发菌株,经500μg/ml NTG诱变处理,获得了具有目的遗传标记寡霉素抗性(O_m^r)和氟乙酸抗性(FEA^r)的突变株TN63和TN115。然后,分别以TN63和TN115为亲株,通过原生质体融合,获得了具有O_m^r+FEA^r标记的融合子FTN9108,其原生质体融合频率为2.6×10^(-3)。确定了目的融合子FTN9108摇瓶发酵的最佳条件,其谷氨酸产率可达8.74g/dl。经连续摇瓶传代10次,发现该融合子是稳定的,具有一定的实用价值。 Using glutamate-producing strain Brevibacterium tianjinese T6-13 as starting strain, mutated by 500μg/ ml NTG, TN63 & TN115 were obtained, which have purpose hereditary marks Omr & EFAr. Using TN63 & TN115 as parents, through the protoplast fusion technique, fusion hybrid FTN9108 which has Omr + FEAr marks was obtained. The frenquency of protoplast fusion is 2. 6×10-3. The best conditions of flask-shaking fermentation of FTN9108 were found. The producing of glutamic acid can be 8. 74g/dl. After 10 generations in the shaking-flask, it keeps the original characteristics and has high applied value.
