摘要
目的:探讨缺血对骨组织超微结构的影响,为吻合血管同种异体骨保存移植提供依据。方法:采用两步冷冻步法,以15%DMSO为低温保护剂,对SD大鼠胫骨进行冷藏缺血和常温缺血实验,时间为1、2、3、4、5、6h和8h,一55℃冷冻温度预处理,液氮保存1周,透射电镜观察。结果:骨缺血1~2h,冷藏组与对照组无明显差异,常温组个别线粒体略有水肿;骨缺血3~4h,冷藏组与常温组仅少数胞浆内线粒体略有肿胀;骨缺血5~6h,冷藏组个别区域骨胶原纤维排列稀疏,线粒体略有水肿。常温组骨细胞水肿,核间隙扩张,骨细胞核有固缩现象;骨缺血至8h,冷藏组骨细胞核间隙水肿扩大,常温组骨细胞核有固缩现象,部分胞质退变,胞膜不完整。结论:缺血8h冷藏的骨组织,其超微结构仍较好,表明冷藏可延长骨组织缺血耐受时限。
Objective: To investigate the effect of the ultra structure of bone tissue of SD rat in different ischemic period. Method: Adopting two frozen methods, the storage duration of ischemia cold group and normal temperature ischemia group of SD rat bone tissue were 1h,2h,3h,4h,5h,6h and 8h respectively. With - 55℃ of preliminary treatment temperature, using 15% DMSO as low temperature protective reagent, the bone tissue were preserved for one week with frozen pretreatment in liquid nitrogen. The tissue was sliced and studied by TEM(transmission electrical microscope). Result:The ischemic bone preserved for 1 to 2h in the cold storage group and contrast group were not obvious different, in the normal temperature groups showed a few mitochondria swelling up. The ischemic bone oreserved for 3 to 4hr,the cold storage groups and normal temperature groups only a few mitochondria in cytoplasm was swelling up. The bone ischemia during 5 to 6hr,the cold storage groups showed the fibre arranged sparseness in some area,the mitochondria was a few swelling up,the normal temperature groups showed dropsy of the bone cell,clearance of nucleus was swelling up, the seasonal striated of fibre not clear. The bone ischemia during 8hr, the bone karyon clearance dropsy swelling up in the cold storage group, the normal temperature groups showed karyopycnosis and some cytoplast degenerate in the bone karyon, the cell membtane is not integrity. Conclusion: This study showed that to ischemia frozen 8h, the ultra structure of bone tissue is in still a good protection, the cold storage of ischemia bone tissue using in this hapel could prolong the duration of bone tissue endurable for ischemia.
