摘要
本文报道用脂肪酸和透明质酸对超氧化物歧化酶进行化学修饰,以改善其稳定性。修饰SOD半衰期为15h,而未修饰SOD仅为5min,活性回收率90~93%,对蛋白水解酶有较强的抵抗力,对热及pH亦具有较强的稳定性。脂肪酸修饰SOD产品比活为4500u/mg,分子量为38000d;透明质酸修饰SOD比活为1013u/mg,分子量为83000d。可用作药用酶、化妆品及食品添加剂。
Abstract: Superoxide dismutase is modified with fatty acid and hyaluronic acid to improve its stability.the resulting conjugate of SOD with fatty acid and hyaluronic acid have a 15h half-life in rats,vs.5min for unmodified SOD. The modified SOD thus prepared retains 90-93% of unmodified SOD activity. The fatty acid-SOD and hyaluronic acid-SOD show greater resistance to pepsin degradation,enhanced heat and pH changes. The fatty acid-SOD is 4500u/mg protein MW 38000d, hyaluronic acid-SOD is 1013u/mg Protein MW 83000d. The preparations may be used as a enzymes for pharmaceutical, cosmetics and food additives.
