盐法结合两步分级沉淀法提取辣木籽蛋白质工艺优化 被引量:1

Optimization of Extraction Process of Moringaoleifera Seed Protein by Salt Method Combined with Two-Step Fractionation Precipitation Method

摘要 以脱脂辣木籽粉为原料,分析辣木籽盐提液中蛋白质的酸碱沉淀条件,在单因素试验基础上结合响应面法优化辣木籽蛋白质的提取工艺条件,通过SDS-PAGE凝胶电泳试验测定该条件下蛋白质提取物的分子量分布情况。结果表明,辣木籽蛋白质的酸沉pH为4.3、碱沉pH为11.0,盐法结合两步分级沉淀法提取辣木籽蛋白质的最佳工艺条件为:NaCl浓度0.2 mol/L、料液比1︰64 g/mL、温度30℃、水浴浸提时间90 min,在此条件下辣木籽蛋白质提取率为65.19%,蛋白质纯度达到86.57%。电泳图显示该蛋白质提取物中酸沉蛋白和碱沉蛋白的分子量在15 kDa、35 kDa、40~60 kDa都有分布,但分子量在25 kDa、35~40 kDa存在较大差异。研究结果为辣木籽蛋白质的进一步开发利用提供参考。 The defatted Moringaoleifera seed powder was used as raw material.The acid-base precipitation condition of Moringaoleifera seed salt extract under different pH was analyzed.The response surface method based on the single factor experiment was used to optimize the extraction condition of Moringaoleifera seed protein.The molecular weight distribution of the Moringaoleifera seed protein extracts was determined by SDS-PAGE gel electrophoresis.The results showed that the acid precipitation pH of Moringaoleifera seed protein was 4.3 and the pH of alkali precipitation was 11.0.The optimum condition for the extraction of Moringaoleifera seed protein by salt method combined with two-step fractionation precipitation method was NaCl concentration 0.2 mol/L,ratio of material to liquid 1︰64 g/mL,temperature 30℃,water bath extraction time 90 min.Under this condition,the extraction rate of Moringaoleifera seed protein was 65.19%,and the protein purity reached86.57%.Electropherogram displayed that the molecular weights of acid precipitation protein and alkali precipitation protein of Moringaoleifera seed protein extracts were distributed at 15 kDa,35 kDa,and 40-60 kDa,but the molecular weights were quite different at 25 kDa and 35-40 kDa between them.The results could provide a reference for further development and utilization of Moringaoleifera seed protein.
