润燥清肺膏成分鉴别及含量测定方法研究 被引量:3

摘要 目的建立润燥清肺膏中桑叶、甘草的鉴别和甘草中甘草酸铵的含量测定方法。方法用薄层色谱法对桑叶、甘草进行定性鉴别;用HPLC法对甘草中的甘草酸铵进行定量检测。色谱条件:色谱柱:Venusil,C18(250 mm×4.6 mm,5μm),以乙腈-0.1%磷酸溶液(36∶64)为流动相;检测波长为250 nm,进样量10μl。结果用建立的薄层鉴别方法可鉴别桑叶、甘草两种药材。甘草酸铵的含量测定在0.056 43~1.128 6μg范围内线性关系良好(r=0.999 8,n=6),平均加样回收率为98.82%,RSD为0.87%(n=6)。结论该鉴别及含量测定方法准确性好、灵敏度高,可用于产品的质量控制,完善了润燥清肺膏的质量标准。 Objective To establish a method for identifying mulberry leaf,liquorice and determining the content of ammonium glycyrrhetate in Runzao Qingfei Ointment. Methods The TLC method was used to identify mulberry leaf and liquorice,HPLC method was used to determine the contents of ammonium glycyrrhetate. The chromatographic conditions were as follows. The venusil C18( 250 × 4. 6 mm,5 μm) column was adopted with the mobile phase of acetonitrile 0. 1% phosphoric acid solution( 36 ∶ 64) at the detection wavelength of 250 nm,the injection volume 10 μl. Results The TLC method can be used for mulberry leaf and liquorice identifying. The sample size of ammonium glycyrrhetate had a good linear relationship with the peak area( r = 0. 999 8,n = 6),within the range of 0. 056 43 ~ 1. 128 6 μg,the average recovery rate was98. 82%,RSD = 0. 87%( n = 6). Conclusion The method is sensitive,simple and accurate,which can be used to control the quality of Runzao Qingfei Ointment. It improves the quality standard.
