通心络对糖尿病肾病大鼠肾组织NO和ET-1的影响 被引量:5

Impact of Tongxinluo on NO and ET- 1 of the Kidney Tissue in the Rats of Diabetic Nephropathy

摘要 目的观察通心络对高脂饲料联合小剂量链脲佐菌素诱导的糖尿病肾病(DN)大鼠肾组织一氧化氮(NO)及内皮素(ET-1)含量的影响,探讨通心络改善DN大鼠肾损害的作用机制。方法采用高脂饲料联合小剂量链脲佐菌素建立DN大鼠模型,将DN大鼠随机分为模型组、缬沙坦组、通心络组各10只,设立10只正常SD大鼠为正常对照组,通心络组给予通心络超微粉0.4 g/(kg·d),缬沙坦组给予缬沙坦10 mg/(kg·d),正常对照组及模型组予等体积蒸馏水,干预16周后检测空腹血糖(FBG),尿微量白蛋白排泄率(UAER)、内生肌酐清除率(Ccr)。于16周末处死大鼠,称量肾重(KW),计算肾重指数(KW/BW),检测肾组织NO及ET-1含量。结果与正常对照组比较,模型组大鼠FBG、UAER、KW及KW/BW、ET-1含量显著升高,差异有统计学意义(P<0.01),Ccr及NO含量显著降低,差异有统计学意义(P<0.01)。与模型组比较,通心络组及缬沙坦组FBG无明显变化,差异无统计学意义(P>0.05),UAER、ET-1、KW及KW/BW显著下降,差异有统计学意义(P<0.05),Ccr及NO含量显著升高,差异有统计学意义(P<0.05)。结论通心络可有效提高糖尿病肾病后期肾组织局部NO水平,降低ET-1含量,改善DN中后期肾小球低滤过,进而保护肾脏,改善肾功能。 Objective To observe the impact of tongxinluo on the levels of nitric oxide( NO) and endothelin( ET- 1) in kidney tissue of the rats with diabetic nephropathy( DN) induced by high- fat forage and small- dose streptozotocin( STZ) and explore the mechanism of tongxinluo on relieving kidney damage in DN rats. Methods The high- fat forage and small- dose STZ were used to prepare DN rat models. The DN rats were randomized into a model group,a valsartan group and a tongxinluo group,10 rats in each one. Additionally,a normal control group was set up with 10 healthy SD rats. In the tongxinluo group,the micropowder of tongxinluo was used,0. 4g / kg d. In the valsartan group,valsartan was used,10 mg / kg d. The distilled water of the equal volume was used in the normal control group and the model group. FBG,UAER and Ccr were detected in 16 weeks after intervention. At the end of the 16 th week,the rats were sacrificed and kidney weight( KW) was measured,KW/BW was calculated and the levels of NO and ET- 1 were detected. Results Compared with the normal control group,the levels of FBG,UAER,KW and KW / BW were increased significantly in the model group,indicating the significant difference( P < 0. 01) and the levels of Ccr and NO were reduced significantly,indicating the significant difference( P < 0. 01). Compared with the normal group,FBG level did not change apparently in the tongxinluo group and valsartan group,without significant difference( P> 0. 05). The levels of UAER,ET- 1,KW and KW / BW were reduced significantly,indicating the significant difference( P < 0. 05). The levels of Ccr and NO were increased significantly,indicating the significant difference( P < 0. 05). Conclusion Tongxinluo effectively increases NO level and reduces ET- 1 level of kidney tissue and relieves the low filtration of glomeruli in DN at the middle and later stages so as to protect the kidney and improve kidney function.
