摘要
目的探讨内皮素-1(ET-1)在早期实验性糖尿病大鼠肾脏中的含量和意义.方法采用免疫组织化学、原位杂交技术检测糖尿病大鼠肾组织ET-1 mRNA含量.结果ET-1 mRNA主要分布于肾小球内皮细胞、系膜细胞、血管平滑肌细胞及肾小管上皮细胞中,髓质高于皮质,糖尿病发生后1~2周肾小球内均明显减少(P<0.05),4周时开始增加(P<0.05),而在髓质中则持续增加.结论ET-1参与早期实验性糖尿病大鼠肾小球高灌注及肾小管间质损伤.
Objective To observe the contents of endothelin-1(ET-1)mRNA in the kidneys of the diabetic rats atearly stages and its significance. Methods Diabetes mellitus was induced by an interperitoneal injection of streptozotocin (55 mg/kg). The contents of ET- 1 were examined by immunohistochenfical staining and then half-quantitative analysis by Mia - 2000 pathology computer image analyzer while the contents of ET - 1 mNA were observed by in situ hybridizational technique. Results The positive signal of ET- 1 mRNA was observed in glomerular endothelial, mesangial, vascular smooth muscles and renal tubular epithelial cells. In glomeruli after 1 - 2 weeks, ET - 1 mRNA decreased significantly compared with the normal control (P〈0.05). But in tubulointerstitial segment of diabetic rats, it increased as compared with the normal controls. Conclusion ET-1 may play an important role in the pathogenesis of glomerular hyperinfusion and tubulointerstitial injury.
