通心络对自发性2型糖尿病KK-Ay小鼠肾小管重吸收功能的影响

摘要 目的观察中药复方通心络对自发性2型糖尿病KK-Ay小鼠肾小管重吸收功能及肾组织megalin和cubilin的影响,探讨中药复方防治糖尿病肾病的作用机制。方法采用KK-Ay小鼠建立糖尿病肾病模型,随机分为模型组、通心络组、缬沙坦组各15只,设C57BL/6J小鼠15只为正常组,给药干预12周后,测定空腹血糖(FPG)、24 h尿微量白蛋白排泄率(UAER)、尿α_1-微球蛋白(α_1-MG)、β_2微球蛋白(β_2-MG)、尿N-乙酰-β-D-葡萄糖苷酶(NAG)含量,采用Western Blot及Realtime PCR方法分别检测肾组织megalin和cubilin的蛋白及基因表达水平。结果与正常组比较,模型组24 h UAER、α_1-MG、β_2-MG、NAG均明显升高(P<0.05),肾组织megalin和cubilin的蛋白和基因表达明显降低(P<0.05);与模型组比较,用药组FPG无明显变化(P>0.05),但24 h UAER、α_1-MG、β_2-MG、NAG等含量明显降低(P<0.05),肾组织megalin和cubilin的蛋白和基因表达上调(P<0.05)。结论通心络可减少2型糖尿病小鼠尿蛋白,改善肾小管重吸收功能,其作用机制可能与上调肾组织中的megalin和cubilin的表达有关。 Objective To observe the effects of tongxiluo on tubular re-absorption as well as the expressions of meglin and cubilin of kidney tissues in KK-Ay mice with type 2 diabetes and explore the effect mechanism of compound herbal medicine in the prevention and treatment of diabetic nephropathy(DN). Methods KK-Ay mice were used to prepare DN models and randomized into a model group,a tongxinluo group and a valsartan group,15 mice in each one. Additionally,15 C57 BL/6 J mice were collected as a normal group. After medication intervention for 12 weeks,the levels of FPG,24 h UAER,α1-MG,β2-MG and NAG were detected. Western blotting and the real-time PCR methods were adopted to detect the protein and gene expressions of megalin and cubilin in kidney tissue separately. Results Compared with the normal group,the levels of 24 h UAER,α1-MG,β2-MG and NAG were increased obviously(P〈0.05) and the protein and gene expressions of megalin and cubilin in kidney tissue were reduced obviously(P〈0.05) in the model group. Compared with the model group,in the medicine group,FPG did not change obviously(P〉0.05),but the levels of24 h UAER,α1-MG,β2-MG and NAG were reduced obviously(P〈0.05) and the protein and gene expressions of megalin and cubilin in kidney tissue were up-regulated(P〈0.05). Conclusion Tongxinluo reduces the urine protein,improves the tubular reabsoption of the mice of type 2 diabetes,which is possibly related to the up-regulation of the expressions of megalin and cubilin in kidney tissue.
