摘要
目的:观察益气清利化瘀方颗粒对慢性肾衰竭(CRF)模型大鼠肾组织形态及转化生长因子-β1(TGF-β1)表达的影响。方法:选择清洁级雄性SD大鼠,采用5/6肾切除方法建立CRF模型。实验设假手术组、模型组、中药组(益气清利化瘀配方颗粒,含生药7.58 g/ml)、西药组(氯沙坦钾片,6.42 g/L);假手术组、模型组均灌胃纯净水,中药组、西药组分别灌胃相应的药物,均2 ml/d。干预60 d后检测血肌酐(SCr)、尿素氮(BUN),计算肌酐清除率(CCr);HE染色观察肾组织病理形态,Western blot法检测TGF-β1蛋白含量。结果:与模型组比较,两药物组SCr、BUN均明显下降(P<0.05,P<0.01),CCr水平显著升高(P<0.01),且中药组对肾功能的改善明显优于西药组(P<0.05)。HE染色显示,两药物组的肾小球纤维化均较模型组有所减轻,且中药组肾小球结构较清晰;与模型组比较,两治疗组皮质、髓质的TGF-β1蛋白表达均较模型组明显减少(P<0.01),且中药组优于西药组(P<0.01)。结论:益气清利化瘀方可改善CRF大鼠肾功能,减轻肾纤维化,下调CRF大鼠TGF-β1表达,其抗肾间质纤维化、改善肾功能的作用可能与抑制肾组织TGF-β1表达及信号转导通路有关。
Objective: To observe the effects of Yiqi Qingli Huayu Recipe on the renal tissue morphology and expression of transfer growth factor-β1( TGF-β1) in rats with chronic renal failure( CRF). Methods: Male SD rats of clean grade were selected. The CRF model was established by 5/6 nephrectomy. The rats were divided into the sham operation group,model group,Chinese medicine( dispensing granules of Yiqi Qingli Huayu Recipe that containing 7. 58 g crude drug/ml) group and western medicine( losartan potassium tablets at concentration of 6.42 g/L) group. The sham operation group and model group were treated with purified water,the Chinese medicine and western medicine group were treated with the corresponding drugs. All the intragastric administration was performed at volume of 2 ml everyday. After treatment for 60 days,the serum creatinine( SCr) and blood urea nitrogen( BUN) were detected,the creatinine clearance rate( CCr) was calculated,the renal histopathology was observed by HE staining,and the protein expression of TGF-β1 was detected by Western blot. Results: Compared with the model group,the levels of SCr and BUN were obviously decreased in Chinese medicine and western medicine group( P<0.05,P<0.01),and the CCr level was significantly increased( P<0.01). The improvement on renal function in the Chinese medicine group was obviously better than that in the western medicine group( P < 0. 05). The HE staining showed that compared with the model group,the glomerular fibrosis was alleviated in Chinese medicine and western medicine group,and the glomerular structure of the Chinese medicine group was clear. Compared with the model group,the protein expression of TGF-β1 in the cortex and medulla was obviously decreased in Chinese medicine and western medicine group( P < 0.01). And the expression levels in the Chinese medicine group were lower than those in the western medicine group( P<0.01). Conclusion: Yiqi Qingli Huayu Recipe can improve the renal function of CRF rats,ease the renal fibrosis and reduce the expression
