摘要
目的观察复方红景天喂食2型糖尿病肾病大鼠尿微量白蛋白(m ALB)和血晚期氧化蛋白产物(AOPPs)的变化,并探究其保护肾脏的作用机制。方法将SPF级8周龄雄性Wistar大鼠50只随机分为正常对照组5只和模型组45只。将成功制作的43只糖尿病肾病模型再随机分为模型对照组10只、小剂量红景天组11只、中剂量红景天组11只、大剂量红景天组11只。小、中、大剂量红景天组分别给予复方红景天3、6、12 g/(kg·d),模型对照组给予等体积生理盐水,连续灌胃4周。治疗前后尾静脉取血检测FBG、FINS、HOMR-IR、TG、TC、AOPPs及12 h尿m ALB水平,并观察大鼠体质量(BW)的变化。结果治疗前,糖尿病肾病模型大鼠BW低于正常对照组,FPG、FINS、HOMR-IR、TG、TC、m ALB、AOPPs水平均高于正常对照组(P<0.05或<0.01)。治疗4周后,小、中、大剂量红景天组BW均高于模型对照组,FPG、AOPPs、m ALB水平低于模型对照组(P均<0.01),小、中、大剂量红景天组BW、AOPPs、m ALB水平比较差异无统计学意义(P均>0.05)。结论复方红景天可能通过降低T2DN大鼠尿m ALB、血AOPPs水平,从而发挥肾脏保护作用。
Objective To observe the changes of microalbuminuria( m ALB) and advanced oxidation protein products( AOPPs) of rats with type 2 diabetic nephropathy( T2DN) by feeding compound rhodiola sachalinensis( Fufang Hongjintian) and to investigate its mechanism of protecting the kidney. Methods Fifty male 8-week-old Wistar rats were randomly divided into the normal control group( A group,n = 5) and model group( n = 45). During the 45 rats,43 rat models with DN were successfully made. Then,they were randomly divided into the model control group( group B,n = 10),low dose compound rhodiola sachalinensis group( group C),medium-dose compound rhodiola sachalinensis group( group D,n= 11),and high-dose compound rhodiola sachalinensis group( group E,n = 11). Rats in the groups C,D and E were treated by 3,6 and 12 g /( kg·d) compound rhodiola sachalinensis and rats in the group B was treated by intragastric administration of the same amount of normal saline for 4 weeks. The FBG,FINS,HOMR-IR,TG,TC,AOPPs and 12-hour m ALB level in the caudal vein of rats were detected before and after treatment and the body weight( BW) was also observed. Results Before treatment,the BW of group B was significantly lower than that of group A,but FPG,FINS,HOMR-IR,TG,TC,m ALB and AOPPs level was higher than that of group A( P < 0. 05 or P < 0. 01). After treatment of4 weeks,the BW of groups C,D and E was higher than that of group B,but FPG,AOPPs and m ALB was lower than that of the group B( all P < 0. 01). No statistical difference was found in FPG,AOPPs and m ALB among groups C,D and E( all P > 0. 05). Conclusion Compound rhodiola sachalinensis( Fufang Hongjingtian) has the protective effect on kidney of T2 DN rats by reducing the levels of m ALB and AOPPs.
