红景天苷对高糖损伤人脐静脉内皮细胞的保护作用研究 被引量:3

摘要 目的观察红景天苷对高糖诱导血管内皮细胞损伤的保护作用及其可能机制。方法实验于2017年2月—2017年11月在青海大学高原医学研究中心实验室进行。体外培养人脐静脉内皮细胞(HUVEC)株贴壁后,以MTS法观察不同浓度葡萄糖对人脐静脉内皮细胞活力的影响,根据糖浓度分组。糖浓度5.5 mmol/L作为正常对照组,糖浓度45 mmol/L作为模型组,以不同红景天苷浓度(1×10^(-5)mol/L、5×10^(-5)mol/L、1×10^(-4)4mol/L、2×10^(-4)4mol/L)作用于模型损伤组,并通过MTS法观察细胞活力,WST-1法测定细胞内超氧化物歧化酶(SOD)活力,TBA法测定细胞内丙二醛(MDA)含量,观察红景天苷对血管内皮细胞损伤的保护作用。结果与模型损伤组比较,细胞培养24 h、48 h,红景天苷5×10^(-5)mol/L、1×10^(-4)4mol/L、2×10^(-4)4mol/L组细胞活力均明显增加(F=5.111、16.330,P<0.01),细胞内MDA含量均明显下降(F=22.567、28.382,P<0.01),细胞内SOD活力均明显增加(F=23.388、82.244,P<0.01);培养48 h,红景天苷1×10^(-5)mol/L组细胞活力与模型损伤组比较显著增加(P=0.04)。结论红景天苷可以降低高糖损伤的HUVEC的氧化应激水平,从而对高糖损伤的血管内皮细胞提供一定的保护作用。 Objective To investigate the protective effect of salidroside on the injury of vascular endothelial cells induced by high glucose and possible mechanism. Methods The HUVECs were cultured in vitro. The group of HUVECs that concentration of glucose was 5.5 mmol/L, which were chosen to be the normal control group, and the group of HUVECs that concentration of glucose was 45 mmol/L, which was selected to be the injury model group, then different concentration of Salidroside were added into injury model group, which were 10^-5 mol/L, 5×10^-5 mol/L, 10^-4 mol/L and 2×10 ^-4 mol/L. Then cell viability was assessed by MTS assay, the level of intracellular superoxide dismutase (SOD) activity were tested in WST-1, and malondialdehyde (MDA) of the different groups were tested in TBA. Results Compared with the model group, when the HUVECs were cultured in 24 hours and 48 hours, Salidroside concentration of 5×10^-5 mol/L, 10 -4 mol/L and 2×10^-4 mol/L had great significance of increasing of the cell viability( F =5.111, P = 0.009; F =16.330, P = 0.00),the level of intracellular MDA had decreased( F =22.567, P =0.00; F =28.382, P = 0.00), and the level of intracellular SOD activity had increased( F = 23.388, P = 0.00; F =82.244, P = 0.00);cultured in 48 hours, compared with the model group, the group that the concentration of Salidroside was 10^-5 mol/L showed more significantly increased of the cell viability( P = 0.04), and then the level of intracellular MDA activity had increased( P = 0.030). Conclusion Salidroside can reduce the level of oxidative stress in HUVEC injury induced by high glucose, thus providing a protective effect on vascular endothelial cells injury induced by high glucose.
