摘要
目的观察人脐带组织间充质干细胞(MSCs)对小鼠B系恶性淋巴瘤和肝癌的趋化作用。方法通过酶切、连接等方法构建p Lenti-f Luc慢病毒表达载体,利用293T细胞包装慢病毒颗粒,并感染人脐带组织华通氏胶来源的MSCs(WJ-MSCs),使其表达萤火虫荧光素酶(f Luc)。分别于NOD/SCID小鼠中建立BJAB淋巴瘤细胞皮下移植模型及BALB/c裸鼠中建立Hep G2肝癌原位移植瘤模型,经尾静脉将MSCs-f Luc注射至荷瘤小鼠体内,采用体内生物发光成像系统观察MSCs-f Luc向肿瘤部位的迁移情况。结果成功构建慢病毒表达载体p Lenti-f Luc,且经慢病毒感染可在WJ-MSCs中稳定表达。成功建立BJAB淋巴瘤细胞皮下移植瘤模型,MSCs-f Luc经小鼠尾静脉注射24 h后选择性地定位于肿瘤部位。成功建立BALB/c裸鼠的Hep G2肝癌皮下原位移植瘤模型,MSCs-f Luc经尾静脉注射2 d后迁移并聚集于肝脏。结论 WJ-MSCs具有向小鼠淋巴瘤和肝癌归巢的能力,此为以MSCs为载体的肿瘤靶向治疗提供了实验基础。
Objective To observe the tropism of human umbilical cord-derived mesenchymal stem cells( MSCs) for malignant lymphoma of B department and liver carcinoma by using in vivo bioluminescent imaging system( IVIS). Methods The lentivirus expression vector p Lenti-f Luc was constructed by restriction enzyme cleavage and linkage. Human umbilical cord Wharton’s Jelly-derived MSCs( WJ-MSCs) were labeled with firefly luciferase( f Luc) by stable transfection with lentivirus which had been packaged in 293 T cells. To demonstrate the tumor tropism of WJ-MSCs,MSC. f Luc were injected intravenously into NOD / SCID mice bearing BJAB lymphoma or BALB / c nude mice which burdened hepatocellular carcinoma in situ with Hep G2 cells,and the migration in vivo was monitored by IVIS. Results The lentivirus expression vector p Lenti-f Luc was successfully constructed,and the expression of f Luc was stable in WJ-MSCs by transfection with lentivirus. The subcutaneous BJAB lymphoma xenograft model was successfully established in NOD / SCID mice,and MSC.f Luc migrated to tumor site after 24 hours by intravenous injection. The Hep G2 orthotopic hepatocarcima model was successfully established in BALB / c nude mice. When it was injected intravenously,MSC. f Luc’s tropism for liver was found 48 hours later. Conclusion WJ-MSCs have the capacity of tumor tropism for lymphoma and hepatocarcima,and it provides basis for tumor-targeting therapy which uses MSCs as the vector.
