摘要
目的:评价吉西他滨对荧光素钾在携带荧光素酶报告基因的乳腺癌细胞株4T1luc胞内的外排动力学的影响。方法:以4T1luc细胞为模型,经不同浓度吉西他滨干预24 h后加入50μg·m L﹣1荧光素钾,应用生物发光成像技术连续动态检测各处理组生物发光信号强度随时间变化过程,计算荧光素钾的外排动力学参数。结果:荧光素钾浓度与生物发光信号光子量呈正相关。随着吉西他滨处理浓度增加,K逐渐增加,AUC逐渐降低(P<0.05)。结论:吉西他滨能够促进乳腺癌细胞胞内荧光素钾的外排动力学过程。
Objective: To evaluate the intervention of gemcitabine( GEM) on efflux kinetics of luciferin potassium in luciferase reporter gene over-expressed 4T1 breast cancer cells( 4T1luc). Methods: Using 4T1 luccells as a model,50 μg·m L- 1luciferin potassium solution was added after 24 h intervention by different concentrations of gemcitabine. Then the bioluminescent signal intensity was detected continuously and dynamically in each treatment group with bioluminescence imaging technology,and the efflux kinetic parameters of luciferin potassium were calculated. Results: The concentration of luciferin potassium was positively related to bioluminescent signal photon.With the increase of gemcitabine concentration,K increased and AUC decreased compared to the control group( P 〈0. 05). Conclusion: Gemcitabine can promote the efflux of luciferin potassium in breast cancer cells.
