摘要
以高效液相色谱 (HPL C)法建立了猪、羊、鸡肝组织中西维因残留量的检测方法。采用 ODS色谱柱 ,二极管阵列检测器 ,甲醇 /水 (6 5 / 35 )为流动相 ,2 2 2 nm检测波长下进行检测。西维因对照品溶液的最低检测限为 0 .1mg/ L,线性范围为 0 .1~ 2 0 mg/ L ,标准曲线回归方程为 :y=- 2 2 4 5 9.83+45 4 6 84 .31x,线性关系良好 (r=0 .9999)。样品以乙腈提取 ,并加入无水硫酸钠和正已烷 ,过滤后去除杂质 ,乙腈层在 4 5~ 5 0℃下水浴氮气吹干 ,残渣用甲醇溶解后 ,进样分析。平均回收率均大于 80 % ,日间和日内变异系数均小于 10 %。符合药物残留痕量分析要求。本方法简便 ,并有很好的选择性、重现性和灵敏度。
A high performance liquid chromatography(HPLC) method using diode array detector(222 nm) for determination of sevin insecticides in swine,goat and chicken liver has been developed.Sevin was separated on Beckman HPLC instrument using YWG 18 (250 mm×4 6 mm,10 μm) column,The mobile phase consisted of a degassed mixture methanol:water(65/35).The flow rate is 1 0 mL/min.Sevin are extracted from liver samples with acetonitrile and evaporated to dryness at 45 50℃ water bath.After evaporation,the residues from the extract are dissolved in methanol and determined by reversed phase high performance liquid chromatography.The limit of detection(LOD) of standard solution is 0 1 μg/mL.When the concentration among 0 1 20 mg/L,the linearity is good.The regression equation is y=- 22 459 83 + 454 684 31 x(r= 0 999 9 ).The average recoveries of sevin was greater than 80% with intra day and inter day coefficients of variations less than 10%.Compared with other methods,it was simple,quick and sensitive.
